Abstract
Vinova loza, kao kultura s dugogodišnjom tradicijom uzgoja u Hrvatskoj i šire, predstavlja jednu od važnijih poljoprivrednih kultura čiji je uzgoj otežan zbog brojnih bakterijskih, gljivičnih, ali i virusnih bolesti. Uvijenost lista vinove loze pridružen virus 3, kao jedan od najrasprostranjenijih i ekonomski najznačajnijih virusa vinove loze, može uzrokovati pad prinosa i do 40%. Prenosi se štitastim ušima, a u svrhu provođenja ovog istraživanja za prijenos je korištena vektorska vrsta Planococcus ficus L. Prijenos je rađen sa zaražene biljke vinove loze na bezvirusne biljke vinove loze te osam korovnih vrsta (Rumex obtusifolius L., Capsella bursa pastoris L., Nicotiana occidentalis H.-M.Wheeler, Nicotiana benthamiana D., Chenopodium album L., Daucus carota L., Matricaria chamomilla L., Galinsoga parviflora Cav). Uspješnost prijenosa provjerena je lančanom reakcijom polimerazom nakon obrnutog prepisivanja. Navedenom metodom potvrđena je uspješnost njegova prijenosa na vinovu lozu od 14,3%, dok prisutnost virusa nije potvrđena ni kod jedne korovne vrste uključene u istraživanje.
Abstract (english)
A grapevine, as a culture with a longstanding tradition of cultivation in Croatia and beyond, represents one of the most significant agricultural crops, whose cultivation is made difficult due to various bacterial, fungal and viral diseases. Grapevine leafroll-associated virus 3, as one of the most prevailing grapevine viruses, can cause a decline in yields up to 40 percent. It is transmitted by mealybugs, and for the purposes of this research vector species, Planococcus ficus L was used. The transfer was made from an infected grapevine plant to healthy grapevine plants and eight weed species (Rumex obtusifolius L., Capsella bursa pastoris L., Nicotiana occidentalis H.-M.Wheeler, Nicotiana benthamiana D., Chenopodium album L., Daucus carota L., Matricaria chamomilla L., Galinsoga parviflora Cav). After the transfer, the samples were analyzed by the PCR method. The efficiency of the transfer to grapevine was 14.3%, while the virus was not successfully transmitted to any weed species.
