Potato is a species that can be infected with various viruses which reduces the yield and quality of tubers. Micro techniques for potato propagation are used to produce healthy plants. Potatoes can be propagated in vitro by micropropagation resulting in the production of rooted plants which are then planted in a greenhouse due to production of healthy mini-tubers; or by microtuberization which produces small microtubers in vitro. Such microtubers can also be used for further production of mini-tubers in a protected area. The aim of this study was therefore to examine the success of micropropagation with the addition of different growth regulators in the medium and the success of microtuberization in different treatments in the traditional potato cultivar 'Brinjak'. Murashige and Skoog medium with the addition of 3% sucrose were used for micropropagation, and the same medium with 8% sucrose was used for microtuberization. Given the quality of the shoots, best root development, and rooting rate, plants micropropagated in medium without growth regulator (HFM-Mt) and medium supplemented with 2.5 mg /l GA3 were likely to achieve the best survival rate after in vitro conditions. Treatment with 2.5 mg /l GA3 + 0.5 mg /l BAP is not recommended for the micropropagation of the traditional cultivar 'Brinjak'due to poor shoot quality. In microtuberization, a higher percentage of shoots with formed microtubers was obtained in the treatment with 4 mg /l kinetin and 1 mg /l 6-benzylaminopurine (KIN4 BAP1) compared to the treatment without growth regulator (HFM-Mt). Also, darkness favorably affected a higher percentage of shoots with formed microtubers. The diameter of the microtubers and the weight of the microtubers were significantly higher in the KIN4 BAP1 treatment on a short day.
