Abstract
Nematode, osim što su paraziti biljaka, čovjeka i životinja, mogu biti i paraziti kukaca. Entomopatogene nematode (EPN) predstavljaju grupu nematoda koje žive u tlu i koje imaju sposobnost parazitacije velikog broja kukaca. Najznačajnije EPN, letalne za štetne kukce, pripadaju porodicama: Steinernematidae, Heterorhabiditidae i Mermithidae. Do sada je opisano više od 70 vrsta iz porodice Steinernematidae i oko 20 vrsta iz porodice Heterorhabditidae. Nematode imaju mutualistički odnos s bakterijama iz dvije porodice Enterobacteriaceae, a zajedno se ponašaju kao moćan insekticidni kompleks. Mogu se uzgajati in vivo ili in vitro, lako se ispuštaju u prirodu, same se obnavljaju, nisu opasne za korisne organizme i prihvatljive su za okoliš. Upravo zbog toga EPN idealne su za biološko suzbijanje.. Dosada u Hrvatskoj, za razliku od većine europskih zemalja, nije utvrđena prisutnost EPN. Iako se radi o korisnim vrstama i prirodnim neprijateljima štetnika, nepoznanica o njihovom postojanju u Hrvatskoj otežava registraciju te uvoz i primjenu pripravaka na osnovu EPN, jer se njihovim unošenjem u okoliš mijenja autohtona fauna nematoda. Cilj ovog diplomskog rada bio je istražiti moguću prisutnost EPN u uzorcima tla prikupljenima u Republici Hrvatskoj. Za potrebe ovoga rada uzgajane su gusjenice voskovog moljca u laboratorijskim uvjetima i klima komori na prikladnim hranidbenim podlogama. Prikupljeno je 25 uzoraka tla na neuznemirenim staništima sa 17 lokaliteta kontinentalne Hrvatske. Postupak izdvajanja EPN proveden je prema metodici „White trap“. U istraživanju nisu izdvojene EPN.
Abstract (english)
Nematodes, mostly known as parasites of plants, humans and animals, are also capable of parasitizing insects. Entomopathogenic nematodes (EPN) represent a group of soil-inhabiting nematodes that parasitize a wide range of insects. These nematodes belong to three families: Steinernematidae, Heterorhabditidae and Mermithidae. Modern research data recognizes more than 70 species of Steinernematidae while there are about 20 described species in the Heterorhabditidae family. Nematodes form a mutualistic partnership with Enterobacteriaceae bacteria thereby creating a potent insecticidal complex. They can be cultured both in vivo and in vitro; are self-replenishing; an are easily released into nature where they pose no harm to the ecosystem. It is because of these qualities that EPN are considered a viable, if not ideal, eco friendly repellent. The presence of EPN in our soiles hasn't been established, thereby preventing the possibility of it's usage, or even import, as means of protection in the biological production of food. The goal oft his thesis was to verify the existence of EPN in sampled soiles from Croatia. For the purposes of this thesis wax moth larvae have been cultivated in controled laboratory conditions. The specimen have been stored in a climate chamber on nutrient media that the wax moth larvae requires. A total of 25 samples have been gathered from 17 various undisturbed locations of continental part of Croatia. The process of EPN isolation was performed using the „White trap“ method. In investigation no EPN have been found.
