Abstract
U ovom radu istraživan je utjecaj inhibitora fotosinteze DCMU-a na učinkovitost fotosustava II u lisnim diskovima japanskog dvornika (Reynoutria japonica Houtt.) uzgojenog u različitim svjetlosnim režimima. U tu svrhu mjerena je fluorescencija klorofila a koristeći uređaj M-PEA. DCMU blokira re-oksidaciju primarnog akceptora QA te uzrokuje inhibiciju fotosintetskog transporta elektrona. Tretman DCMU-om utječe na brže postizanje maksimalnog intenziteta fluorescencije u odnosu na kontrolu, što je vidljivo iz oblika OJIP krivulje: veći nagib početne, OJ faze te smanjena površina iznad OJIP krivulje (Area). To je u konačnici dovelo do smanjenja fotositetske učinkovitosti (PIABS i PItot) tretiranih lisnih diskova.
Abstract (english)
In this work, the effects of DCMU were investigated on photosystem II performance in leaf discs of Japanese knotweed (Reynoutria japonica Houtt.) which were cultivated in different light regimes. In that purpose chlorophyll a fluorescence was measured using the M-PEA analyzer. DCMU blocks re-oxidation of the primary acceptor QA and interrupts the photosynthetic electron chain. The DCMU treatment causes faster achieving of maximum intensity of chlorophyll a fluorescence compared to control, which is visible from the shape of the OJIP transient: initial slope of the OJ phase becomes considerably steeper and surface area above the OJIP transient (Area) was reduced. That lead to decrease of performance index (PIABS, PItot) in treated leaf discs.
