Abstract
Epigenetika je definirana kao nasljedne promjene u aktivnosti i ekspresiji gena koje se događaju bez promjena unutar DNA sekvence. Postoje razni epigenetički mehanizmi koji utječu na regulaciju ekspresije gena. U eukariota, DNA se nalazi unutar jezgre, pakirana s proteinima histonima u nukleosome, glavne jedninice kromatina. Repovi histona su podložni post-translacijskim modifikacijama kao što su metilacija, acetilacija i fosforilacija. Epigenetički procesi kao što su metilacija DNA, modifikacija histona i razni drugi procesi posredovani molekulama RNA, utječu na gensku ekspresiju prvenstveno na razini transkripcije, iako i translacija može također biti epigenetički regulirana. Novija istraživanja se bave povezivanjem utjecaja okoliša i prehrane na epigenetičke mehanizme. Epigenetičke promjene i adaptacije koje se javljaju kao odgovor na uvjete u ranim stadijima razvoja mogu utjecati na rizik obolijevanja i osjetljivost kasnije u životu. Zbog toga razumijevanje epigenetičkih promjena i mehanizama povezanih s odgovorom na okolinu mogu pružiti uvid u osnovne mehanizme početka bolesti kod odraslih.
Abstract (english)
Epigenetics is defined as heritable changes in activity and expression of genes which are happening without the change of the DNA sequence. There are different types of epigenetic mechanisms which affect gene regulation. In eukaryotes , DNA is located within the nucleus,packed with histone proteins. Combined they make chromatine. Main unit of chromatine is nucleosome. Histone tails are subject to post transcriptional modifications such as methylation, acetylation, phosphorylation. Epigenic processes such as DNA methylation, histone modification, and other processes mediated by RNA molecules, can influence gene expression primarily at the transcription level, and also translation level. New researches are trying to understand the connection between the environment and food in regards to epigenetic mechanisms. Epigenetic changes and adaptations which occur during early stages of development may affect the risk of desease development and susceptibility in later stages of life. Because of that understanding epigenetic changes and mechanisms connected to environmental responses can provide insight in basic mechanisms for deseases in adults
