Dataset
Dataset: absorption_and_fluorescence_data.zip, 3.73 MB
Access Condition: Closed access
Description:
The spectroscopic properties of the free-base and Zn(II) complexes of N-methylated pyridinium porphyrins were recorded in methanol and PBS. The data were observed by Martina Mušković under the supervision of Nela Malatesti, PhD. All Excel files contain the raw data of the spectroscopic measurement, and the Origin files contain the analysis of the obtained data. The raw data of the spectroscopic properties after addition of BSA are also included in separate Excel and Origin files. (English)

Dataset: lfp.zip, 37.52 MB
Access Condition: Closed access

Dataset: tcspc.zip, 1.34 MB
Access Condition: Closed access
Description:
The time-correlated single photon counting (TC-SPC) method was used to determine the excited singlet state lifetimes of free base and Zn(II) complexes of porphyrins substituted with an alkyl chain of 7 and 17 carbon atoms. All measurements were performed by Martina Mušković under the supervision of Nikola Basarić, PhD. The attached Origin files contain all the processed data required to record the excited singlet state lifetime as well as the quantum yield of fluorescence of the tested porphyrins in comparison to tetraphenylporphyrin or Zn(II)-tetraphenylporphyrin. (English)

Dataset: singlet_oxygen.zip, 639.83 KB
Access Condition: Closed access
Description:
The singlet oxygen production of free-base and Zn(II) complexes was determined by measuring the photodegradation of two fluorescent dyes, 1,3-diphenylisobenzofuran (DPBF) with methanol as solvent and 9,10-anthracenediyl-bis(methylene)dimalonic acid (ABMDMA) in PBS. Red light (647 nm, 10.7 mW/cm2) and orange light (606 nm, 2 mW/cm2) were used for the irradiation of the DPBF photodegradation measurement and blue light (411 nm, 3.5 mW/cm2) for the photodegradation of ABMDMA. The Excel files contain the raw and processed data of the singlet oxygen production measurements. All measurements were performed by Martina Mušković under the supervision of Nela Malatesti, PhD. (English)

Dataset: lipophylicity.zip, 2.26 MB
Access Condition: Closed access
Description:
Lipophilicity was measured using three methods: modified shake flask method, thin layer chromatography and theoretical calculations using the online program Chemicalize from ChemAxon. The data contains raw Excel files with spectroscopic measurements for calibration curves and with results of shake flask measurements. The GraphPad Prism files contain processed data from shake flask measurements and raw and processed data obtained by thin-layer chromatography. All measurements were performed by Martina Mušković under the supervision of Nela Malatesti. (English)

Dataset: microscopy.zip, 103.63 MB
Access Condition: Closed access
Description:
Fluorescence microscopy was used to determine the intracellular localization of porphyrin. Cells were incubated with porphyrins and DIOC6(3), a fluorescent marker for the detection of ER and mitochondria. All data are original .tif data and data processed in the ImageJ programme. All images were acquired and analysed by Martina Mušković under the supervision of Ivana Ratkaj, PhD. (English)

Dataset: photocytotoxicity.zip, 1.01 MB
Access Condition: Closed access
Description:
The (photo)cytotoxicity of porphyrins was tested using MTT assay on a melanoma (MeWo and A375) and an HDF cell line. Red light (643 nm, 2 mW/cm2) and orange light (606 nm, 2 mW/cm2) were used to irradiate the cells. All data (Excel spreadsheets and GraphPadPrism data) represent raw data obtained after measuring the absorption intensity of the formazan after treatment, as well as processed data with calculated IC50 values and a comparison of proliferation after treatment with 1 uM porphyrin. All data were obtained by Martina Mušković, working under the supervision of Ivana Ratkaj, PhD. (English)

Dataset: cellular_uptake.zip, 737.76 KB
Access Condition: Closed access
Description:
Cellular uptake was determined by measuring fluorescence intensity after incubation of 1 uM porphyrin for up to 24 h at 37 °C and 4 °C on HDF, MeWo and A375 cell line. All Excel files contain raw and processed data, with the concentration (nM) per 1000 cells indicated, as well as GraphPad Prism files with the statistical analysis of the uploaded data. All measurements were performed by Martina Mušković under the supervision of Ivana Ratkaj, PhD. (English)
