Abstract
Humani citomegalovirus (HCMV) je široko rasprostranjeni virus koji, nakon primarne infekcije, uspostavlja latentnu infekciju i doživotno se zadržava u organizmu domaćina. Razlog zbog kojeg organizam domaćina nije u mogućnosti u potpunosti eliminirati virus i na taj način razriješiti infekciju leži u činjenici da genom HCMV-a, kao i drugih citomegalovirusa (CMV-a), kodira za brojne proteine koji suprimiraju imunološki odgovor. Budući da je HCMV visoko specifičan za vrstu i ne može inficirati životinjske stanice, za istraživanje CMV infekcije i imunološkog odgovora na istu se kao model najčešće koristi infekcija miševa mišjim citomegalovirusom (MCMV), čiji genom također kodira za brojne imunoevazine homologne onima za koje kodira genom HCMV-a. Jedan od imunoevazina MCMV-a, protein m04, tijekom infekcije odvodi dio molekula MHC I na površinu inficirane stanice, čime se stanicama NK otežava detekcija i uklanjanje inficiranih stanica. Budući da molekularni mehanizmi kojima protein m04 djeluje na molekule MHC I još uvijek nisu dovoljno razjašnjeni, u ovom je radu konstruiran ekspresijski vektor za ekspresiju proteina m04 u stanicama sisavaca, čime će se omogućiti istraživanje proteina m04 u nativnom obliku izvan konteksta virusne infekcije.
Abstract (english)
Human Cytomegalovirus (HCMV) is a widespread virus that establishes a latent infection after a primary one and is permanently retained in the host organism. The reason the host organism is not able to obliterate the virus and resolve the infection lies in the fact that the HCMV genome and other cytomegaloviruses (CMVs) encode for numerous proteins that suppress the immune response. Considering HCMV is highly species-specific and cannot infect animal cells, the most commonly used model for the CMV infection and immune response research is the murine infection by the murine cytomegalovirus (MCMV) virus, whose genome also encodes for the immunoevasins homologous to those encoded by HCMV genome. One of the MCMV immunoevasins, m04 protein, draws part of the MHC I molecules on the surface of the infected cell during the infection, hence the NK cells are unable to detect and remove the infected cells. An expression vector for expressing m04 proteins in mammalian cells has been constructed in this paper, which will enable the study of protein m04 in native form outside the context of viral infection, since the molecular mechanisms, which the MHC I molecules are affected by m04 protein, have not yet been sufficiently clarified.
