Abstract
Validacija analitičkih metoda ima važnu ulogu u osiguravanju usluge ili proizvoda zadovoljavajuće i pouzdane kvalitete uvjetovane definiranim normama. Ona omogućuje dobivanje korisnih analitičkih podataka visoke točnosti i pouzdanosti nekog mjernog sustava. Cilj ovog rada bio je validirati metodu za kvantitativno određivanje nitrofurantoina tekućinskom kromatografijom visoke djelotvornosti uz detektor s nizom dioda. Da bi saznali kako se nitrofurantoin ponaša u okolišu, ispitivanja su provedena s MilliQ vodom, uzorcima površinskih voda (izvorskoj i morskoj) te sa sintetskom otpadnom vodom. Tijekom validacije HPLC-DAD metode ispitani su ključni parametri: specifičnost / selektivnost, linearnost, osjetljivost, preciznost, istinitost, radno područje, granica kvantifikacije i granica detekcije te stabilnost otopine nitrofurantoina. Rezultati ispitivanja pokazali su da su gotovo svi parametri u skladu s postavljenim kriterijima prihvatljivosti. Prema tome se može zaključiti da je metoda selektivna, linearna i osjetljiva unutar radnog područja, a rezultati mjerenja su točni (precizni i istiniti).
Abstract (english)
Validation of analytical methods plays an important role in ensuring a service or product of satisfactory and reliable quality, conditioned by defined standards. It enables us to obtain useful analytical data of high accuracy and reliability of a measuring system. The aim of this work was to validate the method for the quantification of nitrofurantoin using high performance liquid chromatography with a diode array detector. To find out how nitrofurantoin behaves in the environment, the method was tested using MilliQ water, surface water samples (spring and seawater) and synthetic wastewater. During the validation of the HPLC-DAD method, key parameters were tested: specificity / selectivity, linearity, sensitivity, precision, trueness, working range, limit of quantification and limit of detection, and stability of the solution. Test results showed that almost all parameters were in accordance with the established eligibility criteria. Therefore, it can be concluded that the method is selective, linear and sensitive within the working range, and the measurement results are accurate (precise and true).
