Abstract
Cilj ovog rada jest priprava funkcionalnih nanočestica dopiranih fluorescentnim pH osjetljivim bojilima u svrhu razvoja novih optičkih nanosenzorskih sustava. Kao fluorescentna bojila korišteni su Rhodamin B, lipofilizirano Nile Blue bojilo i 2-piperidinilbenzimidazo[1,2-a]kinolin-6-karbonitril (spoj 1). Kao nosioci navedenih bojila, korištene su silika i polimerne nanočestice. Pripravljene su 3 vrste silika nanočestica (čestice 1, 2 i 3) modificiranom Stöberovom metodom. Čestice 1 pripravljene su miješanjem 30 mL etanola s 1 mL tetraetil ortosilikata (TEOS), 2 mL amonijaka i 0,5 mL vode uz dodatak fluorescentnog bojila Rhodamina B. Čestice 2 su uz sve navedeno sadržavale i (3-aminopropil)trietoksilan (APTES) kao prekursor u sol – gel postupku stvaranja nanočestica, dok su čestice 3 pripravljene na način da je umjesto Rhodamina B (RB) u reakcijsku smjesu dodan derivat Rhodamina B (RBS) funkcionaliziran silanolnim grupama. Pripravljene silika nanočestice okarakterizirane su UV/VIS apsorpcijskom i fluorescencijskom spektrofotometrijom u vodi i etanolu. Također su pretražnom elektronskom mikroskopijom određene veličina i raspodjela nanočestica. Polimerne nanočestice dopirane lipofiliziranim Nile Blue bojilom i bojilom 1 pripravljene su metodom precipitacije otapanjem 50 mg polimera Eudragita® u 25 mL suhog acetona, 1,5 mg lipofiliziranog Nile Blue bojila / bojila 1 te potom naglog dodavanja 150 mL vode. Pripravljene polimene nanočestice dopirane lipofiliziranim Nile Blue bojilom okarakterizirane su UV/VIS apsorpcijskom i fluorescencijskom spektrofotometrijom u vodi i etanolu, dok su polimerne nanočestice dopirane bojilom 1 okarakterizirane u univerzalnom puferu u rasponu pH = 1,0 - 13,0. Također su pretražnom elektronskom mikroskopijom određene veličina i raspodjela nanočestica. U danim eksperimentalnim uvjetima spoj 1 (2-piperidinilbenzimidazo[1,2-a]kinolin-6-karbonitril), zarobljen u polimernim nanočesticama, pokazuje pH odziv koji je uz daljnja istraživanja potencijalno primjenjiv u senzorskim sustavima.
Abstract (english)
The aim of this work was preparation of functional nanoparticles doped with fluorescent pH-sensitive dyes in order to develop novel optical nanosensor systems. Rhodamine B, lyophilized Nile Blue and 2-piperidinilbenzimidazo [1,2-a] quinoline-6-carbonitrile (dye 1) were used as fluorescent dyes. Silica and polymer nanoparticles were used as dye carriers. Three kind of silica nanoparticles were prepared (particles 1, 2 and 3) by modified Stöber method. Particles 1 were prepared by mixing 30 mL of ethanol with 1 mL of tetraethyl orthosilicate (TEOS), 2 mL of ammonia and 0.5 mL of water with the addition of Rhodamine B. Particles 2 had all of the above with addition of (3-Aminopropyl)triethoxysilane (APTES), while particles 3 were prepared in a way that instead of adding Rhodamine B (RB), modified Rhodamine B (RBS) was added to the reaction mixture. Prepared silica nanoparticles were characterized by UV / VIS absorption and fluorescence spectroscopy in water and ethanol. In addition, size and distribution of nanoparticles were determined by scanning electron microscopy. Polymeric nanoparticles doped with lipophilised Nile Blue dye and dye 1 were prepared by the precipitation method by dissolving 50 mg of polymer Eudragit® in 25 mL of dry acetone, 1.5 mg of lyophilised Nile Blue dye / dye 1 an the rapid addition of 150 mL of water. Prepared polymeric nanoparticles doped with Nile Blue dye were characterized by UV / VIS absorption and fluorescence spectroscopy in water and ethanol, whereas polymeric nanoparticles doped with dye 1 were characterized in water and ethanol as well as in universal buffer at pH = 1,0 – 13,0. In addition, size and distribution of nanoparticles were determined by scanning electron microscopy. It was found that compound 1 (2-piperidinilbenzimidazo [1,2-a] quinoline-6-carbonitrile), trapped in a polymeric nanoparticle, shows the pH response which is, along additional research, potentially usable in sensor systems.
