Abstract
Validacija analitičke metode je nužan korak u osiguravanju kvalitete u analitičkoj kemiji. To je postupak koji obuhvaća definiranje zahtjeva, eksperimentalno ispitivanje izvedbenih karakteristika metode te davanje izjave o zadovoljavanju postavljenih zahtjeva. Kako bi se u potpunosti validirao sustav moraju se ispitati sve izvedbene karakteristike metode, a one su: specifičnost/selektivnost, linearnost, točnost, istinitost, preciznost, granica detekcije i kvantifikacije, radno područje i robusnost. Cilj ovog rada bio je validirati metodu za određivanje metoklopramida te ispitati karakteristike i na temelju rezultata zaključiti zadovoljava li analitička metoda kriterije prihvatljivosti. Analiza je provedena HPLC-MS/MS metodom na otopinama metoklopramida u MilliQ, izvorskoj i morskoj vodi, te su ispitane sve izvedbene karakteristike metode. Rezultati istraživanja su pokazali kako je metoda zadovoljila sve kriterije prihvatljivosti osim kriterija istinitosti za otopinu morske vode najmanje koncentracije. Iz toga možemo zaključiti kako je metoda prihvatljiva za određivanje metoklopramida.
Abstract (english)
Validation of the analytical method is a necessary step in ensuring quality in analytical chemistry. It is a procedure that includes defining requirements, experimental testing of method performance characteristics and giving a statement on meeting those requirements. In order to fully validate the system, all performance characteristics of the method must be examined, and they are: specificity/selectivity, linearity, accuracy, trueness, precision, limit of detection and quantitation, working range and ruggedness. The aim of this study was to validate the method for determination of metoclopramide and to examine the characteristics and, based on the results to conclude whether the analytical method meets the acceptance criteria. The analysis was performed by HPLC-MS/MS method on solutions of metoclopramide in MilliQ, spring and sea water and all performance characteristics of the method were examined. The result of the study showed that the method met all the acceptance criteria except the trueness criteria for the sea water solution of the lowest concentration. From this we can conclude that the method is acceptable for the determination of metoclopramide.
