Abstract
Cilj ovog rada je različitim tehnikama shvatiti što se događa na nivou membrane kada dođe do interakcije flavonoida s membranom. Infracrvenom spektroskopijom (IR) je istražen utjecaj flavonola miricetina (MCE) i miricitrina (MCI) na temperaturu prijelaza, Tm, modelnog membranskog lipida 1,2-dimiristoil-sn-glicero-3-fosfokolina (DMPC) iz stanja gela u stanje tekućih kristala. Mikroskopijom atomskih sila (AFM) istražene su orijentacije lipida u membrani, a pomoću tehnike dinamičko raspršenje svjetlosti (DLS) istražen je naboj liposoma. Multilamelarni liposomi snimani su u transmisiji kako bi se odredila Tm, dok su spektri hidratiziranih višestrukih lipidnih dvosloja snimani apsorpcijsko-refleksijskom spektroskopijom na horizontalnoj površini (HATR) kako bi se odredio smještaj flavonola u dvosloju. Rezultati dobiveni mjerenjima pokazuju da flavonoli snizuju Tm DMPC-a što je posljedica njihove ugradnje u dvosloj. MCE ulazi dublje u lipidni dvosloj, dok MCI zaostaje na hidrofilnoj površini dvosloja.
Abstract (english)
The aim of this research is to understand what happens on membrane level when it comes to intereaction with flavonoids by using various techniques. IR spectroscopy was used to study effects of flavonols myricetin (MCE) and myricitrin (MCI) on temperature of phase transition, Tm, from gel phase to liquid crystalline phase of model membrane lipid 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC). The orientation of lipids in the mambrane was observed using Atomic force microscopy (AFM) and charge of liposoms by using Dynamic light scattering (DLS). Multilamellar liposomes were recorded in transmission in order to determine Tm. Spectra of the hydrated multibilayers were recorded by Horizontal Attenuated Total Reflection (HATR) technique to determine their position in the bilayer. Results indicate that flavonols interact with DMPC and that their incorporation in the bilayer causes lowering of Tm. MCE goes deeper into the lipid bilayer, while MCI is more on the hydrophilic surface of the bilayer.
