Abstract
U ovom radu određivana je kinetika oksidoredukcije Cbz-N-3-aminopropanola u Cbz-N-3-aminopropanal katalizirana enzimom alkohol dehidrogenazom iz konjske jetre (HLADH). Ova reakcija je sastavni dio kaskadne reakcije koja se sastoji od opisane oksidacije alkohola u aldehid, potom aldolne adicije dihidroksiacetona (DHA) na pripadajući aldehid koja je katalizirana enzimom D-fruktoza-6-fosfat aldolazom (FSA) kako bi se dobio odgovarajući aldolni produkt, odnosno (3S,4R)–6–[(benziloksikarbonil)amino]–5,6– dideoksiheks–2-uloza. Ova kaskadna reakcija također uključuje i regeneraciju koenzima NAD+ uz NADH oksidazu (NOX) izoliranu iz Lactococcus lactis, te je kao takva uzeta u obzir pri ispitivanju kinetike. Provedena su eksperimentalna ispitivanja enzimske aktivnosti u prethodno opisanoj reakciji oksidoredukcije, te su na temelju dobivenih podataka i procijenjeni kinetički parametri. Razvijen je matematički model kaskadne reakcije, te su provedene simulacije pomoću programskog paketa SCIENTIST kako bi se ispitao utjecaj pojedine komponente u reakciji na konačnu koncentraciju aldolnog produkta.
Abstract (english)
Kinetics of oxidoreduction of Cbz-N-3-aminopropanol to Cbz-N-3-aminopropanal catalyzed by horse liver alcohol dehydrogenase (HLADH) was studied in this work. This reaction is a constituent part of a cascade reaction composed of the described oxidation from alcohol to aldehyde, following with the aldol addition of dihydroxyacetone (DHA) to the corresponding aldehyde, catalyzed by D-fructose-6-phosphate aldolase (FSA) for the production of aldol product, (3S, 4R)-6-[(benzyloxycarbonyl)amino]-5,6-dideoxyhex-2-ulose. This cascade reaction also contains a coenzyme regeneration of NAD+ catalyzed by NADH oxidase (NOX) isolated from Lactococcus lactis and was taken into consideration in the study of kinetics. Enzyme activity was studied in the previously described oxidoreduction reaction. The kinetic parameters were estimated from the data obtained in the conducted experiment. Mathematical model for the cascade reaction was developed and simulated by using the software package SCIENTIST. The influence of concentrations of different components of the reaction system to the final aldol product concentration was simulated.
