Abstract
Razvojem industrije i tehnologije, počela se razvijati i farmaceutska industrija, koja je danas jedan od vodećih sektora. Lijekovi koji se proizvode postoje u raznim formama, od kojih su najčešće forme u obliku praha i tableta, proizvedene procesom kristalizacije. Procesom kristalizacije nastaje nova, kristalna faza, uglavnom iz otopine. Kod takvih kristalizacija potrebno je obratiti pozornost na otapalo koje se koristi. Osim što otapalo mora otapati određenu tvar i biti kemijski čisto, u obzir se moraju uzeti i ekonomičnost, sigurnost i stabilnost otapala. Nakon kristalizacije otapala se izdvajaju u obliku matičnih lugova, točnije smjese otapala, aktivne tvari i drugih nečistoća koje zaostanu za vrijeme procesa kristalizacije. Za regeneraciju otapala koriste se razni separacijski procesi, od kojih je destilacija najčešća. U ovom je radu istražena mogućnost regeneracije organskih otapala (korištenih u PLIVI, Istraživanje i Razvoj) različitim metodama separacije. Literaturno su istražene različite mogućnosti separacije kao npr. destilacije, ekstrakcije i pervaporacije ili njihovih kombinacija. Najveći naglasak je dan na destilaciju i to šaržnu destilaciju pri atmosferskom ili sniženom tlaku, zatim destilaciju uz pomoćnu komponentu (tzv. entrainer – etilen glikol ili propilen glikol), te ekstraktivnu ili Dean Stark destilaciju. Cilj je bio provesti regeneraciju tetrahidrofurana (THF) iz matičnih lugova proizvodnje djelatnih farmaceutskih tvari, te ispitati koji proces omogućava regeneraciju THF-a do željene čistoće, uzimajući u obzir i ekonomičnost. Provedeni su šaržni eksperimenti destilacije matičnih lugova u koloni sa sitastim pliticama sa 10 teorijskih plitica, te u šaržnom reaktoru sa jednostavnom kolonom i Dean – Stark nastavkom. Svi eksperimenti su opisani matematičkim modelima i simulacijama pomoću programa ChemCAD i DynoChem. U obzir se uzela mogućnost zamjene organskih otapala ekološki prihvatljivim otapalima (kao npr. 2-metiltetrahidrofuran).
Abstract (english)
With the growth of industry and technology, pharmaceutical industry also started to advance and today it became one of the leading industries. Pharmaceuticals are produced in many forms, typically in bulk dosage forms and tablets, mainly by crystallization. In the process of crystallization new phase is produced from the solution. In that type of crystallization it is necessary to choose the best solvent. Not only does the solvent need to dissolve the right compound, it must be chemically clean, economical, safe and stabile. After crystallization, solvents are separated in form of mother liquor, that is, mixture of solvents, active compound and other impurities that are left in from the process of crystallization. Distillation is the main separation process for regeneration of solvent. In this paper the possibility of regeneration of organic solvents (used in PLIVA, R&D) with different separation processes, such as distillation, extraction and pervaporation, or its combinations, to targeted specified purity is explored. In the process of regeneration many types of distillation are used: batch distillation at atmospheric and reduced pressure, distillation with entrainer (ethylene glycol, propylene glycol) and extractive or Dean – Stark distillation. The main objective was to regenerate THF from mother liquor from production of active pharmaceutical ingredients and to investigate the possibility of the regeneration to target specification as well as economy. Experiments of distillation of mother liquors were conducted in column with 10 theoretical sieve trays and batch reactor with simple column and Dean – Stark trap. Experimental data were correlated with mathematical models using software’s ChemCAD and Dynochem. Also, the substitution of organic solvents with green solvents (such as 2-Me THF) was explored.
