Abstract Centaurea triumfettii All. (eng. Squarrose knapweed) pripada porodici Asteracea i zeljasta je višegodišnja biljka trajnica. Cilj ovog diplomskog rada bio je izolirati eterično ulje iz osušenog biljnog materijala C. triumfettii, metodom hidrodestilacije, korištenjem modificirane aparature po Clavenger-u. Kemijski sastav eteričnog ulja C. triumfetii utvrđen je instrumentalnom tehnikom plinska kromatografija-spektrometrija masa (GC/MS). U eteričnom ulju C. triumfettii identificirano je ukupno 30 komponenti koje čine 87,2% ukupnog ulja. Spojevi s najvećim prisustvom su heksadekanska kiselina (11,1%), spatulenol (10,8%), longifolen (8,8%) i germakren D (8,4%), zatim slijede aromadendren oksid (6,0%), linoleinska kiselina (5,3%) i α-kadinol (4,5%). Kako bismo dobili informaciju o spojevima koji se nalaze u sastavu eteričnog ulja C. triumfetii i njihovom potencijalnom antimikrobnom učinku; prikupljeni su do sada istraženi podaci o antimikrobnom djelovanju eteričnih ulja Centaurea vrsta i obrađeni statistički, korištenjem poznatih informatičkih alata za obradu podataka. Sveukupno je obuhvaćeno 30 različitih eteričnih ulja od 27 Centaurea vrsta koja su testirana na gram- pozitivnim, gram-negativnim bakterijama te gljivicama. Fokus našeg istraživanja bio je na bakterijama E. coli i S. aureus. Dominanti kemijski spojevi iz eteričnog ulja C. triumfettii, heksadekanska kiselina, longifolen, aromadendren oksid i linoleinska kiselina, ne nalaze na popisu spojeva koji imaju značajnu korelaciju sa antimikrobnim učinkom. Germakren D, spatulenol i α-kadinol se pojavljuju sa značajnom korelacijom na antimikrobni učinak, no skup podataka iz kojih je izašla ova informacija je malen i dobivene rezultate treba uzeti s oprezom. Navedene korelacije kemijskih spojeva koji se nalaze u sastavu eteričnih ulja Centaurea vrsta, koje se povezuju sa antimikrobnim učinkom, mogu služiti kao smjernica u istraživanju, te nikako ne možemo sa sigurnošću tvrditi da su navedeni spojevi zaista odgovorni za antimikrobni učinak dok ne ispitamo njihovo djelovanje u obliku čistih spojeva.
Abstract (english) Centaurea triumfettii All. (Squarrose knapweed), is a herbaceous perennial plant who belongs to Asteracea family. The aim of this thesis was to isolate the essential oil from the dried plant material C. triumfettii, using the hydrodistillation method and modified Clavenger apparatus. Chemical composition was analyzed by coupled system gas chromatography-mass spectrometry (GC / MS). In the C. triumfettii essential oil, a total of 30 components were identified, being 87.2% of the total oil. The compounds with the highest yield were hexadecanoic acid (11.1%), spathulenol (10.8%), longifolene (8.8%) and germacrene D (8.4%), followed by aromadendrene oxide (6.0%). linoleic acid (5.3%) and α-cadinol (4.5%). In order to obtain information on the chemical compounds in the C. triumfetii essential oil and their antimicrobial effect; the data on the antimicrobial activity of Centaurea essential oils have been investigated to date and processed statistically using known IT tools for data processing. In total, 30 different essential oils of 27 Centaurea species have been tested for gram-positive, gram-negative bacteria and fungi. Focus of our research was on microbes E. coli and S. aureus. The dominant compounds of C. triumfettii essential oil, hexadecanoic acid, longifolene, aromadendrene oxide and linoleic acid are not on the list of chemical compounds that have a significant correlation with the antimicrobial effect. Germacrene D, spatulenol, and α-cadinol appear to have a significant correlation to the antimicrobial effect, but the data set from which this information was extracted is small and the results should be taken with caution. The above correlations of the chemical compounds found in the Centaurea species essential oils, which are associated with antimicrobial activity, can serve as a guideline for research, and we cannot say with certainty that these compounds are indeed responsible for the antimicrobial effect until we examine their effect in form pure compounds.
