Abstract Imidakloprid je insekticid koji se koristi za zaštitu biljaka od insekata. Najčešće dolazi u obliku formulacije pod nazivom Confidor. Imidakloprid spada u grupu neonikotionida što znači da djeluje agonistički na nikotinske receptore u živčanom sustavu. Iako mu je primarni mehanizam djelovanja poznat, ne može se isključiti činjenica da možda postoji neki drugi put djelovanja. Prema tome, potrebno je istražiti je li kod imidakloprida, uz djelovanje na nikotinske receptore, prisutno i sekundarno djelovanje. Za takva istraživanja pogodni su model organizmi kao što je kvasac Saccharomyces cerevisiae. Kvasci dijele mnoge metaboličke procese s ljudskom stanicom, stoga se rezultati djelovanja imidakloprida na stanicu kvasca mogu prenijeti i na ljudsku stanicu. U ovom radu istraženo je potencijalno djelovanje Confidora na metabolizam lipida u stanicama kvasca S. cerevisiae. Kvasci su tretiriani Confidorom te su iz njih izolirani ukupni lipidi (metodom po Folchu) i mitohondriji. Nadalje, lipidi su razdvojeni po frakcijama (neutralni lipidi, fosfolipidi, slobodne masne kiseline, sterolni esteri, triacilgliceroli, steroli, diacilgliceroli i monoacilgliceroli) ekstrakcijom na krutoj fazi, SPE (solid phase extraction). Lipidi pojedinih frakcija su zatim metilirani te analizirani plinskom kromatografijom. Rezultati su pokazali da Confidor djeluje na kvasce, smanjujući njihovu vitalnost. Između kontrolnih uzoraka i uzoraka kvasaca tretiranih Confidorom postoje razlike u sastavu masnih kiselina. Također, razlike postoje i u udjelima pojedinih frakcija lipida pa se tako udjeli sterola, triacilglicerola i monoacilglicerola značajno razlikuju između kontrolnog uzorka i uzorka kvasca tretiranog Confidorom. Može se zaključiti da Confidor djeluje na lipide kvasca S. cerevisiae mijenjajući njihov sadržaj i sastav.
Abstract (english) Imidacloprid is a pesticide used to protect the plants from insects. Usually it comes in a formulation called Confidor. Imidacloprid belongs to the group of neonicotinoids, meaning it acts agonistically on nicotine receptors in the nervous system. Although its primary mechanism of action is well known, there is a possibility that there may be some other mechanisms. Therefore, it is necessary to investigate whether there is a secondary mechanism present in imidacloprid acting, along with the impact on nicotine receptors. Model organisms such as yeast Saccharomyces cerevisiae are suitable for such studies. Yeasts are similar in many metabolic processes with the human, so the results of imidacloprid impact on the yeast cells can be transfered to the human cells. This work investigates the impact of Confidor on S. cerevisiae lipid metabolism. The yeast was treated with Confidor and the total lipids (using Folch method) and mitochondria was isolated. Furthermore, lipids were separated on fractions (neutral lipids, phospholipids, free fatty acids, sterol esters, triacylglycerols, sterols, diacylglycerols and monoacylglycerols) by solid phase extraction (SPE). The lipids of individual fractions were then methylated and analyzed by gas chromatography. The results have shown that Confidor has an impact on yeasts, reducing its vitality. Between control samples and samples of yeast treated with Confidor there are differences in the composition of fatty acids. Also, there are differences in quantity of individual lipid fractions, so that sterol, triacylglycerol and monoacylglycerol levels differ significantly between the control sample and the yeast treated with Confidor. It can be concluded that Confidor has an effect on lipids of yeast S. cerevisiae which can be seen in changes of their composition and content.
