Abstract (english)
Introduction Arf proteins are small GTPases that function as molecular switches and cycle between their active (GTP) and inactive (GDP) form. Their activation is mediated by guanine nucleotide exchange factors (GEFs), and their inactivation is facilitated by the GTPase-activating proteins (GAPs). We have shown earlier that Arf proteins have important role in the development of murine cytomegalovirus (MCMV) assembly compartment (AC) in the early phase of infection. In the late phase of infection, late-phase viral proteins accumulate in the outer part of AC, which is one of the preconditions for the production of the new virions. Aims The aim of this study was to determine intracellular localization of Arf proteins and their GEFs in the late phase of MCMV infection and to determine if they accumulate in the same compartment as late-stage MCMV viral proteins. Materials and methods Balb 3T3 were infected with recombinant murine cytomegalovirus Δm138-MCMV and at 30 or 48 hours post infection (30 hpi or 48 hpi) intracellular localization of Arfs and Arf GEFs was analyzed by confocal microscopy. Arf proteins and their GEFs were colocalized with different late-phase viral proteins, including m55, m74 and m25. Results In the late phase of MCMV infection no significant colocalization of Arf proteins or Arf GEFs with late-phase MCMV viral proteins was observed. The only exception is Arf3 that showed partial colocalization with late-phase MCMV viral proteins. Conclusion Arf proteins, with exception of Arf3, and their GEFs are excluded from outer part of MCMV in the late phase of MCMV infection.
