Abstract
O fetalnim mastocitima ljudskog sjemenika u dostupnoj literaturi praktički nema podataka. Hipoteza ove disertacije je da broj mastocita postupno raste s gestacijskom dobi fetusa te da se razvoj ovih stanica podudara spovećanjemvolumena cjelokupnog sjemenika i njegovih struktura. Stoga je cilj ovog rada bio prikazati mastocite u parenhimu fetalnog sjemenika odgovarajućim histološkim tehnikama te istražiti razvojnu dinamiku ovih stanica tijekom intrauterinog razvoja.U radu je korišten fetalni materijal dobiven uobičajenom pedopatološkom sekcijom 39 spontano pobačenih plodova u dobi od 15. do 36. tjedna gestacije. Tijekom analize korištene su kvalitativne i kvantitativne histološke metode. U ranijim fazama razvoja (15.-29. tjedan) mastociti su smješteni isključivo unutar tunike albugineje fetalnog sjemenika. Tek od 30. tjedna gestacije pa sve do kraja ispitivanog razdoblja (36. tjedan) mastocitinaseljavaju i rahlo vezivo tunike vaskuloze te vezivne pregrade/rahlo vezivo u dubini parenhima fetalne muške spolne žlijezde. Ukupan broj mastocita postupno raste tijekom ispitivanog razdoblja razvoja. Rezultati kvantitativne histološke analize ukazuju na podudarnost u razvojnoj dinamici mastocita ipovećanja volumena sjemenika,spolnih tračaka i intersticija. Za razliku od toga promjer spolnih tračaka se bitno ne mijenja. Pretpostavlja se da je razvoj mastocita u fetalnom ljudskom sjemeniku uvjetovan različitim parakrinim čimbenicima stanica koje izgrađuju intersticij i spolne tračke. Ovi čimbenici, ukoliko se izoliraju, mogli bi u budućnosti biti primjenjeni u terapiji neplodnosti muškarca, osobito u slučajevima neopstruktivne azoospermije.
Abstract (english)
In the literature there are no available data about mast cells in the human fetal testis. The hypothesis of this study was that the number of mast cells increases with gestational age of the foetus, coinciding with the increase of volumes, sex cords and interstitium. The aim was to visualize mast cells within the parenchyma of the foetal testis by histological staining methods and investigate the development of these cells during the intrauterine development of the whole testis and its structures. The material was obtained during the routine paedopathological autopsy of 39 spontaneously aborted/stillborn foetuses between 15 and 36 gestational weeks. Qualitative and quantitative analysis was done. During gestational weeks 15-29, mast cells were exclusively located within the tunica albuginea. After 30th gestational week mast cells, in addition to tunica albuginea, mast cells populated the loose connective tissue of the tunica vasculosa and the connective tissue of septa/interstitium within the parenchyma. The total number of mast cells increased over the investigated period. Quantitative sterological analysis showed that the development of mast cells coincided with the increase of the volumes of the testis, sex cords and the interstitium. In contrast to these results, diameter of the sex cords remains similar. The development of mast cells in the foetal testis is probably regulated by paracrine factors secreted by cells of the interstitium and sex cords. Once isolated, they could present possible targets for treatment of the male infertility.
