Abstract
Tumori testisa, poznati kao tumori zametnih stanica testisa (engl. testicular germ cell tumors, TGCT), predstavljaju najučestalije neoplazme koje zahvaćaju muškarce u reproduktivnoj dobi. Čine 95% svih slučajeva tumora testisa te su klasificirani kao heterogena skupina neoplazmi, koja se klasificira na seminome i neseminome. TGCT se često javljaju kao miješani tumori s raznolikim histološkim podtipovima: embrionalnog karcinoma, teratoma, tumora žumanjčane vreće i koriokarcinoma. Radi precizne dijagnostike, terapije TGCT-a i detekcije prisutnosti specifičnih histoloških podtipova, u kliničkoj praksi koriste se serumski i tkivni biomarkeri. Glavni cilj ovog istraživanja bio je istražiti specifičnost ekspresije antigena DPPA3 unutar pojedinih komponenti TGCT-a te ispitati njegov potencijal kao dijagnostičkog biomarkera za razlikovanje pojedinih podtipova neseminomski tumora zametnih stanica testisa. Za provedbu istraživanja korištena su tkiva pacijenata, pri čemu je 38 uzoraka TGCT-a, dok je 20 tkiva bez neoplastičnih promjena. Ukupno 58 bioloških tkiva testa. U istraživanju su primijenjene metode histološkog bojanja, imunohistokemije i statističke analize podataka. Klinički patolog je proveo kvantifikaciju signala u pojedinačnim komponentama TGCT-a, nakon čega su podaci statistički obrađeni. Rezultati istraživanja su pokazali značajnu razinu ekspresije antigena DPPA3 u stanicama embrionalnog karcinoma. Dobiveni rezultati istraživanja ukazuju na potencijal antigena DPPA3 kao dijagnostičkog markera za razlikovanje embrionalnog karcinoma od ostalih podtipova TGCT-a. Ova saznanja doprinose boljem razumijevanju bioloških karakteristika TGCT-a i otvaraju perspektivu za daljnja istraživanja s ciljem identifikacije novih tkivnih i serumskih biomarkera kao i terapijskih pristupa za ovu skupinu tumora.
Abstract (english)
Testicular cancer, known as testicular germ cell tumors (TGCT), is the most common neoplasm affecting men of reproductive age. They make up 95% of all cases of testicular tumors and are classified as a heterogeneous group of neoplasms divided into seminomas and non seminomas. TGCTs often present as mixed tumors with diverse histological subtypes: embryonal carcinoma, teratoma, choriocarcinoma, and yolk sac tumor. In clinical practice, serum and tissue biomarkers are used for precise diagnosis, treatment, and detection of the specific histological subtypes of TGCT. This research aimed to investigate the specificity of the DPPA3 antigen within individual components of TGCT and to examine its potential as a diagnostic biomarker for differentiating individual subtypes of non seminomatous germ cell tumors of the testis. Patient tissues were used in this research, where the number of TGCT samples was 38, while the number of tissues without neoplastic changes was 20. Histological staining, immunohistochemistry, and statistical data analysis methods were used in the research. A clinical pathologist quantified the signal in each component of TGCT, after which the data were statistically processed. The research results showed a significant level of DPPA3 antigen expression in embryonic carcinoma cells. The obtained research results indicate the potential of the DPPA3 antigen as a diagnostic marker for distinguishing embryonal carcinoma from other subtypes of TGCT. These findings contribute to a better understanding of the biological characteristics of TGCT and open the perspective for further research to identify new biomarkers and therapeutic approaches for this group of tumors.
