Abstract
Biomarker je bilo koja molekula koja uzrokuje promjenu fiziološkog stanja pojedinca na bolest,terapiju ili toksine. U tom pogledu, autoantitijela povezana s karcinomima su odlični kandidati za detektiranje karcinomskih biomarkera koji se pojavljuju kao reakcija imunološkog sustava na tumor. Stvaranje autoantitijela koja se pojavljuju prema autolognim karcinomskim antigenima nije statično i mjerljivo je različitim analizama. Autoantitijela su izrazito stabilna u humanom serumu i dugo perzistiraju nakon uklanjanja antigena. To je njihova izrazita prednost te se koriste kao potencijalni markeri u tjelesnim tekućinama za detekciju karcinomskih antigena. Za razliku od autoantitijela, karcinomski antigeni imaju kratko poluvrijeme raspada. Zbog toga, stabilnost autoantitijela je bitna u razvoju laboratorijskih testova. Glavna uloga pronalaska autoantitijela je u tome da se razlikuje fiziološko stanje od patološkog, prvenstveno u ranim asimptomatskim fazama karcinoma. Probir biomarkera mogao bi poboljšati dijagnostičku vrijednost medicinskog statusa, testova i ostalih procedura. Ostali testovi probira kao što su mamografija i kolonoskopija pomažu u pronalasku inicijalnih znakova karcinoma. Istraživanje autoantitijela je bitno da bi se utvrdila bolest u inicijalnoj fazi. U otkrivanju biomarkera su razvijene razne strategije. Osjetljivost i specifičnost testova mora biti dovoljno visoka da bi se autoantitijela upotrijebila za skrining. Potrebno je daljnje istraživanje autoantitijela i tehnika koja bi ih otkrivale. To bi moglo doprinijeti ranoj dijagnostici karcinoma i većem uspjehu terapije.
Abstract (english)
A bimarker is any molecule that indicates an alteration of the physiological state of an individual relevant to disease state, treatment and toxins. In this field, autoantibodies are beign considered as excellant cantidates for cancer markers as they represent response of the immune system. The generation of these autoantibodies in response to autologus cellular antigenes is not static and it is measurable by analyses. Autoantibodies are extremely stable in serum samples and are know to persist for extended times after removal of the antigenic factor. This is a distinct advantage for their use and are potential markers for detection of carcinoma in human fluids. Cancer antigens have limited half-life. Indeed, the stability of autoantibodies is imortant for development of laboratory tests. The main objective of autoantibodies discovery is to identify molecular markers capable of discriminating the „healthy“ state from the „disease“ state preferentially in the early asymptomatic stages. Biomarker screening test may enhance the diagnostic value of physical examination, medical tests or other procedures. Cancer screening procedures such as mammography or screening colonoscopy serve in finding initial signs of cancer. Different strategies have been developed in discovering biomarkers. The sensitivity and the sensibility of screening test must be high for the detection of the autoantibodies. Developing strategies for autoantibodies are needed. That may contribute to early cancer detection and improvement of therapy results.
