Abstract
CILJ ISTRAŽIVANJA: Ciljevi ovog diplomskog rada bili su odrediti prisutnost i intenzitet imunohistokemijskog izražaja HIF-1α na parafinskim rezovima bioptičkih uzoraka pacijenata s primarnim melanomom ,odrediti prisutnost i intenzitet imunohistokemijskog izražaja HIF-1α na parafinskim rezovima bioptičkih uzoraka pacijenata s metastatskim melanomom, odrediti lokaciju (boji li se jezgra i/ili citoplazma) imunohistokemijskog izražaja HIF-1α u uzorcima pacijenata s primarnim odnosno metastatskim melanomom. MATERIJALI I METODE: Istraživanje je provedeno na 20 bioptičkih uzoraka melanoma uklopljenih u parafin bolesnika koji su liječeni u klinici za onkologiju KBC-a Split. Uvidom u medicinsku dokumentaciju izdvojeno je 10 pacijenata s primarnim melanomom i 10 pacijenata s metastatskim melanomom . Od svakog je pacijenta, iz arhive Kliničkog odjela za patologiju, Kliničkog zavoda za patologiju, sudsku medicinu i citologiju KBC-a Split, izdvojen parafinski blok s uzorkom tumorskog tkiva (primarnog/metastatskog melanoma). Određena je prisutnost i intenzitet imunohistokemijskog izražaja HIF-1α u tumorskim stanicama pacijenata s primarnim melanomom te u pacijenata s metastatskim melanomom. REZULTATI: Rezultati ovog rada pokazuju da je HIF-1α prisutan u svim uzorcima primarnog te metastatskog melanoma. Intenzitet imunohistokemijskog izražaja HIF-1α je u uzorcima primarnog melanoma blag (50%) ili umjeren (50%), dok je u uzorcima metastatskog melanoma blag (30%), umjeran (50%) ili jak (20%). U obje skupine uzoraka odnosno i u uzorcima primarnog kao i u uzorcima metastatskog melanoma izražaj HIF-1α je bio u jednakom postotku smješten samo u citoplazmi (60%), odnosono u jezgri i citoplazmi(40%). ZAKLJUČAK: HIF-1α je prisutan u svim uzorcima primarnog te metastatskog melanoma. Intenzitet imunohistokemijskog izražaja je veći u uzorcima metastatskog melanoma (blag/umjeren/jak) u odnosu na uzorke primarnog melanoma(blag/umjeren). S obzirom na navedeno , možemo zaključiti kako bi HIF-1α zaista mogao imati važnu ulogu u procesu rasta i napredovanja melanoma te kako bi se možda mogao koristiti kao prediktor pojavnosti metastaza melanoma u regionalne limfne čvorove. Ujedno HIF-1α bi u budućnosti mogao postati ciljana meta u terapiji melanoma.
Abstract (english)
OBJECTIVES: The aim of this study was to determine immunohistochemical expression of HIF-1α and its intensity in bioptic samples of patients with primary melanoma and patients with metastatic melanoma and to determine the localization of the expression of HIF-1α. EXAMINEES AND METHODS: The study was done on 20 paraffin sections of biopsy samples of patients with malignant melanoma who were threated at the Department of Oncology, University Hospital of Split. We choosed 10 patients with primary melanoma and 10 patients with metastatic melanoma whose histopathological material was taken from the archives of the Clinical Department of Pathology, Clinical Institute of Pathology, Forensic Medicine and Citology, Clinical Hospital Split. RESULTS: Results of this study indicate that Hif-1α was present in all samples of primary melanoma and metastatic melanoma, and that there was positive immunohistochemical expression. Intensity of immunohistochemical expression of HIF-1α in samples of primary melanoma was mild (50%) or moderate (50%), while in metastatic melanoma samples was mild (30%), moderate (50%) or stong (20%). Localisation of immunohistochemical expression of HIF-1α was same in both groups of samples (primary and metastatic melanoma samples); citoplasm (60%),and citoplasm and nucleus (40%). CONCLUSION: Hif-1α was present in all samples of primary melanoma and metastatic melanoma, and there was positive immunohistochemical expression. There was strong upregulation of intensity of HIF-1α expression in metastatic melanoma samples in comparison to primary melanoma samples, that suggested that HIF-1α might be an important factor for malignant melanoma growth and in the future might be predictor for regional lymph node metastasis. Also HIF-1α could be target molecule for malignant melanoma therapy.
