Abstract
Cilj istraživanja: Cilj ovog istraživanja je ispitati ulogu genetske podloge ljudi koja ih čini sklonima nastanku meningitisa, na temelju podatka iz povijesti bolesti o bolničkom liječenju meningitisa te rezultata određivanja njihovih genotipova. Materijali i metode: Korišteni su podatci iz projekta 10.001 Dalmatinac, i to s otoka Visa, otoka Korčule i grada Splita. Ukupno je u istraživanje bilo uključeno 3.007 ispitanika, od kojih je za njih 26 postojao podatak o bolničkom liječenju meningitisa u njihovoj povijesti bolesti. Koristili smo dob i spol ispitanika, s ciljem otkrivanja genetske podloge za nastanak meningitisa. Ispitanici su uz ove podatke dali i uzorak krvi koji je korišten za ekstrakciju DNA. Ispitanici su nakon toga bili genotipizirani, korištenjem dva različita panela. U analizi podataka korišten je pristup kandidatnog gena, u kojem se otprije poznati biljezi koriste kako bi se njihova vrijednost vrednovala u drugoj populaciji. U analizi je korišteno ukupno 47 biljega jednog nukleotida, koji su bili pronađeni u pet ranije objavljenih istraživanja. Rezultati: Rezultati su ukazali na granično značajan ishod za dva biljega jednog nukleotida, koji su oba pripadali genu SP110. Ostali kandidatni geni nisu bili značajno povezani s ishodom. Zaključak: Rezultati ovog istraživanja ukazuju na to da bi gen SP110 mogao imati vrlo važnu ulogu u nastanku meningitisa, neovisno o patogenom uzročniku. Ovi rezultati govore u prilog osobnom riziku za nastanak meningitisa, što predstavlja osnovicu za razvoj individualiziranog pristupa određivanju rizika za nastanak meningitisa.
Abstract (english)
Objectives: The aim of this study was to investigate the genetic background of meningitis risk, based on their medical history and analysis of genetic characteristics of the host. Materials and methods: We used the data from the 10,001 Dalmatian project, originating from the island of Vis, the island of Korčula and the city of Split. There were 3,007 subjects, with 26 of them having meningitis in their medical history. Study used demographic parameters (age and gender) with the aim of discovering the genetic basis for the occurrence of meningitis. Subjects gave a blood sample that was used for the extraction of DNA. Subjects were subsequently genotyped, using two different panels. In analyzing the data, the access candidate gene was used, where previously known markers are used in order to evaluate their value in the other population. The analysis used a total of 47 single nucleotide markers that were found in the five previously published studies. Results: The results showed borderline significance for two single-nucleotide markers, which both belonged to the gene SP110. Other candidate genes were not significantly associated with the outcome. Conclusion: The results of this study indicate that the gene SP110 could have a very important role in the development of meningitis, regardless of the pathogens. These results support the idea of personal risk assessment, which is the basis for the development of an individualized approach to determining the risk of meningitis.
