Abstract
Kvasac Pichia stipitis je zanimljiv zbog sposobnosti unutarstaničnog akumuliranja (S)-adenozil-L-metionina (SAMe). Mutanti dobiveni ultraljubičastim svjetlom i rezistentni na nistatin imaju promijenjen sastav sterola u staničnoj membrani, odnosno mogu nakupljati SAMe koji se ne iskoristi u biosintezi ergosterola. Cilj ovog rada bio je usporediti karakteristike (potrošnja glukoze, prirast biomase, sadržaj SAMe u biomasi kvasca, tolerancija na etanol) divljeg soja P. stipitis i mutiranog soja M12 (UV mutant NisR) tijekom uzgoja na tresilici u kemijski definiranim podlogama. Uspoređujući ova dva soja primijećen je znatno brži rast odnosno potrošnja glukoze divljeg soja kvasca P. stipitis u odnosu na mutirani soj M12. Postignut je znatno veći sadržaj SAMe u biomasi mutiranog soja M12 uzgojenog u mediju uz dodatak 1,5 g/L D,L-metionina u usporedbi s divljim sojem P. stipitis, dok je u mediju bez dodatka D,L-metionina uočena neznatna razlika u količini SAMe prisutnog u biomasi ispitivanih sojeva kvasca. Utvrđena je i niža tolerancija na etanol mutiranog soja M12 u usporedbi s divljim sojem P. stipitis.
Abstract (english)
Yeast Pichia stipitis is interesting because of its ability to intracellular concentrations of (S)-adenosyl-L-methionine. Mutants obtained by ultraviolet radiation and resistant to nystatin have altered sterol composition within cellular membrane and can accumulate SAM that is not utilaze in the ergosterol biosynthesis. The aim of this work was to compare the characteristics (consumption of glucose, biomass growth, content of SAM in yeast biomass, tolerance to ethanol) of P.stipitis wild strain and mutant M12 (obtained by UV and resistant to nystatin) during cultivation in a chemically defined media in a shaker. Comparing these two strains, much more rapid growth and consumption of glucose were observed in wild yeast strain P. stipitis than in mutant M12. The amount of SAM in the biomass of mutant M12 grown in medium with the addition of 1.5 g/L D,L-methionine was significantly higher compared to the wild strain P. stipitis, while in the medium without the addition of D, L-methionine only slight differences in the amount of SAM present in the tested strains of yeast were revealed. A lower tolerance to ethanol was determined in the mutant M12 compared to the wild strain of P. stipitis.
