Abstract
Kulture životinjskih stanica upotrebljavaju se u svim područjima biotehničkih znanosti. Prilikom uzgoja kulture najčešće je nužan dodatak seruma koji stanici osigurava sve tvari potrebne za rast, metabolizam i proliferaciju. Razvojem procesa s kulturama životinjskih stanica sve su veći zahtjevi za smanjenjem upotrebe seruma i razvoja medija bez dodatka seruma. Danas su razvijeni različiti tipovi adaptacije na medij bez dodatka seruma, a kako je svaka kultura različita, stanice je potrebno postupno prilagoĎavati smanjenjem koncentracije seruma i zamjenom seruma sa sastojcima koji su potrebni za rast i održavanje kulture. Cilj ovog rada bio je ispitati rast stanične linije CCO u mediju bez seruma postupnim smanjivanjem udjela seruma od 10 % do 0,1 %. Postupak prilagodbe kao i odabir medija bez seruma su se pokazali uspješnim budući da je postignut dobar rast CCO stanica pri 0,1 % seruma.
Abstract (english)
The animal cell cultures are used in every segment of biotechnology. To achive a successful development of a cell line, it is very important to add serum, which supplies the cell culture with substances necessary for its growh, metabolism and proliferation. With the evolution of animal cell culture there is an increasing demand for reducing the usage of serum and for developing serum-free mediums. Scientists have developed different types of adaptation to serum-free medium. Since every culture is different, cells must be slowly adjusted to the serum-free medium by reducing the concentration of the serum and by replacing it with substances necessary for the growth of the cell line.. The main goal of this work was to compare growth of CCO cell line in a medium without serum while progressively reducing the share of the serum from 10 % do 0,1 %. Process of adaptation, as well as a choice of serum-free medium, was very successful since the growth of the CCO cell line at 0,1 % of serum was satisfaying.
