Abstract
U ovom radu izolirana je plazmidna DNA iz kvasca Saccharomyces cerevisiae (soj BY4741), koji je prethodno transformiran plazmidom pYES-SCE. Prisutnost plazmida dokazana je gel-elektroforezom, pri čemu su zabilježene dvije konformacije plazmida (superzavojnica i otvoreni krug). Plazmidna DNA dodatno je podvrgnuta restrikcijskoj analizi s dva restrikcijska enzima (HindIII and ClaI), te su vrpce linearizirane DNA, dobivene gel-elektroforezom, bile u skladu s restrikcijskom mapom plazmida pYES-SCE. Ovaj je rezultat potvrdio da je soj kvasca Saccharomyces cerevisiae bio uspješno transformiran plazmidom pYES-SCE.
Abstract (english)
In this study the plasmid DNA was isolated from the yeast Saccharomyces cerevisiae (strain BY4741) previously transformed with the plasmid pYES-SCE. The presence of the plasmid was confirmed by gel-electrophoresis and two conformations (supercoil and open circle) of the circular plasmid were observed. Additionally, the plasmid DNA was subjected to restriction analysis with two restriction enzymes (HindIII and ClaI) and the bands of the cut DNA obtained by gel electrophoresis corresponded to the restriction map of the plasmid pYES-SCE. This result confirmed that the strain of the yeast Saccharomyces cerevisiae was successfully transformed with the plasmid pYES-SCE.
