Abstract
Za protein Scw4, koji se nalazi u staničnoj stijenci kvasca Saccharomyces cerevisiae, još uvijek nije poznato sudjeluje li u mejozi i sporulaciji te koja je njegova uloga u tim procesima. Taj protein nekovalentno i kovalentno je vezan za staničnu stijenku, pretpostavlja se da ima glukanaznu aktivnost i stoga vjerojatno sudjeluje u organizaciji glukana. Stanična stijenka, ekstracelularna je organela koja se sastoji od β-1,3 i β-1,6-glukana, hitina i manoproteina. Ulaskom u mejozu i sporulaciju dolazi do brojnih morfoloških i genetičkih promjena stanične stijenke, primjerice smanjenja ili povećanja ekspresije određenih proteina. Cilj ovog rada je utvrditi prisutnost proteina Scw4 tijekom mejoze i sporulacije. Dodatkom hemaglutininske (HA) oznake na C terminalni kraj proteina Scw4 omogućeno je praćenje ekspresije specifičnim anti-HA antitijelima nakon izolacije proteina. Obilježavanje i detekcija proteina uspješno su provedene, a Scw4 je u SDS-ekstraktu prisutan u mejozi i sporulaciji. Ovaj rad doprinosi razumijevanju uloge Scw4 proteina u staničnoj stijenci kvasca.
Abstract (english)
It is still not known if Scw4 protein in yeast Saccharomyces cerevisiae cell wall plays role in meiosis and sporulation. That protein is non-covalently and covalently attached to cell wall, and it's assumed that it has glucanase activity and probably participates in glucan organization. Cell wall is the extracellular organelle which consists of β-1,3 and β-1,6-glucan, chitin and mannoproteins. Meiosis and sporulation results in many morphological and genetical changes of cell wall, for example, decrease or increase of certain protein expression. The goal of this work is to determine presence of Scw4 during meiosis and sporulation. After protein isolation, it is enabled to track Scw4 expression with specific anti-HA antibodies by adding hemagglutinin (HA) tag to C-terminal residue of Scw4. Tagging and detection are successfully conducted and in SDS-extract Scw4 is present in meiosis and sporulation. This science work contributes in understanding of the role of Scw4 in cell wall.
