Abstract
Kondenzirana struktura kromatina djeluje represivno na sve biološke procese u eukariotskim stanicama koji se odvijaju na DNA, uključujući transkripciju, te je stoga remodeliranje strukture kromatina promotorske regije preduvjet za aktivaciju transkripcije inducibilnih gena. Promotor gena PHO5 kvasca S. cerevisiae pokazao se kao izvrstan modelni sustav za istraživanje mehanizma remodeliranja kromatina pri aktivaciji transkripcije. U ovome radu ispitan je učinak inaktivacije kromatin-remodelirajućeg kompleksa RSC na indukciju gena PHO5 u stanicama mutanata rsc kod kojih je taj gen pod kontrolom inačica promotora nižeg praga indukcije i različitog aktivacijskog potencijala u odnosu na nativni promotor. Praćenjem kinetike indukcije pokazano je da je indukcija ovih inačica promotora PHO5 manje ovisna o aktivnosti kompleksa RSC nego indukcija nativnog promotora, ali je znatno niža od razine indukcije u stanicama s aktivnim kompleksom RSC.
Abstract (english)
In eukaryotic cells, the condensed structure of chromatin exerts a repressive effect on all biological processes involving DNA such as transcription and, therefore, chromatin remodeling of the promoter region is a prerequisite for the activation of transcription of inducible genes. Yeast PHO5 promoter is regarded as an exceptional model system for studying the mechanism of chromatin remodeling and subsequent activation of transcription. This study examined the effect of the ablation of the chromatin-remodeling complex RSC on PHO5 induction in yeast rsc mutants which had the PHO5 gene under the control of promoter variants with a lower induction threshold and different activation potentials than the wild type promoter. The results showed that the induction of such PHO5 promoter variants is less dependent on the activity of RSC complex than the induction of the wild type promoter, but the induction level is still significantly lower than in the cells with active RSC complex.
