Abstract
Provedena je analiza biokatalitičke sinteze (S)-2-hidroksipropiofenona iz benzaldehida i acetaldehida katalizirana enzimom benzoilformijat dekarboksilazom (BFD) iz bakterije Pseudomonas putida u različitim tipovima reaktora (kotlasti, ultrafiltracijski enzimski membranski i mikroreaktorima). Ispitana je kinetika reakcije u kotlastom reaktoru i mikroreaktorima, te efektivna difuzivnost benzaldehida, acetaldehida i BFD enzima u mikroreaktoru. Na temelju rezultata postavljeni su matematički modeli. Usporedbom volumnih produktivnosti različitih tipova reaktora pokazalo se da su mikroreaktori (posebno mikroreaktor s mikromiješalima) bolji izbor pri provoñenju biokatalitičke sinteze (S)-2- hidroksipropiofenona. Ispitivan je i sustav za recirkulaciju enzima u mikroreaktoru i na temelju dobivenih rezultata može se zaključiti da navedeni sustav u mikroreaktorima ima veliki potencijal za biokatalitičku sintezu (S)-2-hidroksipropiofenona. Takoñer je ispitivana enantioselektivna redukcija (S)-2- hidroksipropiofenona do (1S,2S)-1-fenilpropan-1,2-diola katalizirana enzimom alkohol dehidrogenazom iz bakterije Lactobacillus brevis i (1R,2S)-1-fenilpropan-1,2-diola katalizirana enzimom glicerol dehidrogenazom iz bakterije Cellulomonas sp. Ispitivanja su provedena u kotlastom reaktoru bez i sa regeneracijom koenzima. Bez regeneracije koenzima nije bilo moguće postići potpunu konverziju supstrata, dok je potpuna konverzija supstrata postignuta u sustavu s regeneracijom koenzima u kojem je korišten i enzim formijat dehidrogenaza iz kvasca Candida boidinii kao regeneracijski enzim. Odreñeni su kinetički parametri reakcija i na osnovi rezultata postavljeni su matematički modeli procesa.
Abstract (english)
Biocatalytic synthesis of (S)-2-hydrohypropiophenone form benzaldehyde and acetaldehyde catalyzed by benzoylformate decarboxylase enzyme (BFD) isolated from Pseudomonas putida was performed in different reactor types (batch, ultrafiltration enzyme membrane and microreactors). Kinetics of the reaction was determined for batch reactor and microreactor and effective diffusivities of benzaldehyde, acetaldehyde and BFD enzyme in microreactor. Based on experimental data mathematical models were proposed. By comparing the results for volume productivity for different reactor types microreactors (especialy microreactor with micromixers) are more suitable for the biocatalytic synthesis of (S)-2- hydrohypropiophenone. System for microreactors wich recirculates the enzyme was tested and based on the experimental data it could be concluded that use of the enzyme recirculation in microreactors has a great potential for biocatalytic syntheses. Also enantioselective reduction of (S)-2-hydrohypropiophenone to (1S,2S)-1- phenylpropane-1,2-diol catalyzed by alcohol dehydrogenase from Lactobacillus brevis and (1R,2S)-1- phenylpropane-1,2-diol catalyzed by glycerol dehydrogenase from Cellulomonas sp. was analyzed. The experiments were done in batch reactor without and with coenzyme regeneration. Without coenzyme regeneration it was not possible to achieve full conversion of substrate, but with coenzyme regeneration using formate dehydrogenase from Candida boidinii the conversion was 100 %. For these reactions kinetic parameters were determined and mathematical models were proposed and validated.
