Abstract
Gotovo svi stanični procesi, počevši od ekspresije gena do sinteze i razgradnje proteina mogu biti pod utjecajem prehrane i životnog stila. Nutritivne i nenutritivne komponente na kompleksan način utječu na metaboličke promjene u stanici. Primjena metodologije matematičkog modeliranja omogućuje dobivanje detaljnog uvida u složene procese u stanici na brz, jednostavan i ponovljiv način. U ovom radu proveden je dizajn i simulacija modela metabolizma glikolize u programskom paketu CellDesigner. Ispitan je utjecaj različitih početnih koncentracija glukoze te smanjenih aktivnosti glukokinaze, fosfofruktokinaze i piruvat kinaze povezanih sa šećernom bolesti na metabolizam glukoze. Primjena računalnog programa CellDesigner pokazala se kao vrlo koristan alat za analizu specifičnog metabolizma glikolize kod oboljelih od šećerne bolesti s obzirom da je moguća vizualna interpretacija utjecaja promjena koncentracija i brzina reakcija pojedinih enzima.
Abstract (english)
Almost all cellular processes, from gene expression to proteins synthesis and degradation can be affected with nutrition and life style. Nutritive and non-nutritive food components affect metabolic changes in the cell in a complex way. The application of the mathematical modelling methodology ensures detailed insight into complex processes in the cell in a fast, simple and repetitive manner. In this paper, the design and simulation of the glycolysis metabolism model were carried out in the CellDesigner software package. The effect of various initial glucose concentrations on glucose metabolism and the effect of reduced glucokinase activity, phosphofructokinase and pyruvate kinase associated with diabetes mellitus has been studied. The CellDesigner software has been shown to be a very useful tool for the analysis of specific glycolysis metabolism in diabetic patients since it is possible to visually interpretate the effect of the concentration change and reaction rate of certain enzymes.
