Abstract
Velika količina lignoceluloznog otpada, kojeg čine uglavnom celuloza, hemiceluloza i lignin, nastaju u šumarstvu, poljoprivredi i prehrambenoj industriji. Lignoceluloza je kompleksna obnovljiva sirovina koja se može prevesti u razne visokovrijedne biotehnološke proizvode kao što su bioetanol, bioplin i biogoriva, ali se može koristiti i za proizvodnju mikrobne biomase te lignocelulolitičkih enzima koji su od sve veće industrijske važnosti. U ovom radu provedena je proizvodnja lignocelulolitičkih enzima uzgojem gljive bijelog truljenja Dichomitus squalens na otpadnim drvnim ostacima mekog (lipa) i tvrdog (hrast) drva te su izmjerene enzimske aktivnosti proizvedenih ekstracelularnih enzima i izračunate koncentracije razgradnih produkata nakon provedenog uzgoja. Izmjerene su i uspoređene aktivnosti enzima lakaze, mangan peroksidaze, celobioza dehidrogenaze, ksilanaze, arabinaze, celulaze, karboksimetil celulaze te pektinaze. Cilj eksperimenta bio je utvrditi utjecaj različitih izvora ugljika na uzgoj Dichomitus squalens za postizanje maksimalne aktivnosti enzima.
Abstract (english)
Huge amounts of lignocellulosic waste, consisting mainly of cellulose, hemicellulose and lignin, are generated through forestry, agriculture and food industry. Lignocellulose is the complex renewable raw material that can potentially be converted into various valuable biotechnological products such as bioethanol, biogas and biofuels, but it can also be used to produce microbial biomass and lignocellulolytic enzymes that are growing in industrial importance. In this research, the production of lignocellulolytic enzymes was performed by growing the white-rot fungus Dichomitus squalens on waste wood residues of soft (linden) and hard (oak) wood, and the enzymatic activities of the produced extracellular enzymes together with the concentrations of degradation products were measured. The activities of the enzymes laccase, manganese peroxidase, cellobiose dehydrogenase, xylanase, arabinase, cellulase, carboxymethyl cellulase and pectinase were measured and afterwards compared. The aim of this study was to determine the influence of different carbon sources on the cultivation of Dichomitus squalens for maximum enzyme activity.
