Abstract
Ljekovita biljka medvjetka se od davnina koristi za liječenje i ublažavanje tegoba izazvanih urinarnim infekcijama. Budući da još nisu utvrđene interakcije između bakterija i epitela mokraćnog mjehura kada su izloženi djelovanju aktivnih tvari iz medvjetke, cilj ovog rada bio je utvrditi citotoksični i antioksidacijski učinak hidrokinona na bakteriju Escherichia coli i humane stanice raka mokraćnog mjehura (T24). U radu je istražen i utjecaj hidrokinona na promjenu adhezije ove bakterije na stanične linije T24. Rezultati su pokazali toksični učinak hidrokinona na bakteriju u koncentraciji koja odgovara sadržaju hidrokinona u jednoj šalici napitka pripravljenog od medvjetke, pri čemu je dokazana veća citotoksičnost hidrokinona kada su bakterije tretirane 1h u odnosu na 2h. Utvrđen je učinak iste koncentracije hidrokinona na smanjenje adhezije E. coli na stanice T24 kao i visok antioksidativni učinak. Doza hidrokinona u šalici napitka, uz pretpostavku da sav arbutin tijekom konzumacije metabolizira u hidrokinon, predstavlja izrazito citotoksičnu koncentraciju za T24 stanice gdje u uvjetima s 24-satnim oporavkom smanjuje njihovo preživljenje ispod 20% u odnosu na kontrolu.
Abstract (english)
The medical plant bearberry has been used for healing and alleviating the problems caused by urinary infections since distant past. Since interaction between bacteria and bladder epithelium when exposed to activity of active substance from bearberry has not been established yet, the aim of this study was to determine the cytotoxic and antioxidant effect of hydroquinone on E. coli and human bladder cancer cells (T24). Also, ability of hydroquinone to alter the adhesion of E. coli to T24 cells was investigated. The results showed the toxicity of the hydroquinone to the bacterium at a concentration corresponding to the hydroquinone content in a single cup of beverage, whereby was demonstrated the higher cytotoxicity of the hydroquinone when the bacteria were treated for 1 h over 2 h. The effect of the same hydroquinone concentration on the reduction of E. coli adhesion to T24 cells and the high antioxidant effect were determined. The dosage of hydroquinone in a cup of beverage assuming that all the arbutin during metabolism is metabolised into hydroquinone, is a highly cytotoxic for T24 cells where 24-hour recovery conditions reduce their survival below 20 %.
