Abstract
Karcinom dojke je najčešća maligna bolest koja se javlja kod žena i jedan je od najčešćih karcinoma u svijetu. Njegov razvoj često je potaknut prekomjernom aktivnošću signalnih puteva u stanici, među koje spada RAS-RAF-MAPK signalni put. Transmembranski receptor ERBB2 uključen je u RAS-RAF-MAPK signalni put i provodi signal od izvanstaničnog čimbenika rasta do unutrašnjosti stanice. Istraživanjima je dokazano da amplifikacija i prekomjerna ekspresija HER2 gena, gena koji kodira za ERBB2 receptor, te onkogene mutacije RAS i RAF izoformi posljedično dovode do razvoja različitih podtipova karcinoma dojke, naročito HER2-pozitivnog i TNBC podtipa. Tretiranjem RAS-RAF-MAPK signalnog puta anti-ERBB2 protutijelima te RAF i MEK inhibitorima otvara se mogućnost liječenja karcinoma dojke.
Abstract (english)
Breast cancer is the most common malignancy that occurs in women and is one of the most common cancers in the world. Its development is driven by excessive activity of signalling pathways in the cell, including the RAS-RAF-MAPK signalling pathway. The ERBB2 transmembrane receptor is involved in the RAS-RAF-MAPK signalling pathway and carries the signal from the extracellular growth factor to the cell interior. Studies have shown that amplification and overexpression of HER2 gene, a gene encoding the ERBB2 receptor, and oncogenic mutations in RAS and RAF isoforms consequently lead to the development of different subtypes of breast cancer, especially HER2-positive and TNBC subtypes. Treatment of the RAS-RAF-MAPK signalling pathway with ERBB2-antibodies and RAF and MEK inhibitors opens the possibility of treating breast cancer.
