Abstract
Danas su u primjeni mnoge metode koje se osnivaju na metodi PCR (Polymerase Chain Reaction, lančana reakcija polimerazom), a jedna od njih je RAPD-PCR ili RAPD (Random Amplification of Polymorphic DNA, metoda slučajnog umnažanja polimorfne DNA). Reakcijske otopine za provođenje metode PCR i metode RAPD sadrže iste komponente, ali uvjeti provođenja reakcije su različiti. Metoda PCR se koristi za umnažanje točno određenog dijela DNA iz nekog genoma, a metodom RAPD umnaža se veći broj različitih fragmenata DNA. Stoga se usporedbom RAPD profila (umnoženih fragmenata) može donijeti zaključak o srodnosti analiziranih organizama odnosno postojanju polimorfizama u njihovim genomima. Glavne prednosti metode su niska cijena i kratko vrijeme potrebno za provođenje analize, a mana je što jako male promjene u uvjetima reakcije mogu značajno utjecati na reproducibilnost rezultata analize.
Abstract (english)
Many methods based on PCR (Polymerase Chain Reaction) are being used nowadays. One of them is RAPD-PCR or RAPD (Random Amplification of Polymorphic DNA). PCR and RAPD reaction solutions consist of same components but conditions in which the reaction is carried out differ in these two methods. PCR is used to amplify a specific part of a DNA from a genome, while RAPD enables the amplification of many different DNA fragments. By comparing RAPD profiles (amplified products), a conclusion about similarity between different organisms or the existence of polymorphisms in their genomes can be made. Main advantages of the method are low price and short analysis time, while its disadvantage is the significant influence of reaction conditions on the reproducibility of results.
