Abstract
Cilj ovog rada bio je ispitati kakav učinak imaju različite koncentracije antioksidansa poput resveratrola i kurkumina te njihovih novosintetiziranih ferocenskih konjugata na razinu ROS-a i vijabilnost stanica u CHO (Chinese Hamster Ovary) - K1 staničnoj liniji. U tu svrhu proveden je test za detekciju ROS-a te MTT test za procjenu vijabilnosti stanica. Pri tretmanu stanica ferocenskim konjugatom resveratrola (RF) razina ROS-a bila je najmanja u odnosu na preostala tri ispitana spoja te je stoga upravo taj bioorganometalni spoj odabran za određivanje mogućeg protektivnog učinka pri tretmanu stanica induktorima oksidacijskog stresa kao što su tert-butil hidroperoksid (TBHP) i kongener polikloriranih bifenila – PCB 77. RF je u svim ispitanim koncentracijama (1; 2,5 i 20 μM) značajno smanjio ROS induciran 50 μM TBHP-om. Također, određena je niža razina ROS-a u onim stanicama koje su bile predinkubirane RF-om, a zatim izložene 75 μM PCB-u 77 u odnosu na stanice izložene samo PCB-u 77, no u ovom slučaju učinak nije bio statistički signifikantan.
Abstract (english)
The aim of this study was to investigate the effect of different concentrations of antioxidants such as resveratrol and curcumin and their novel synthesized ferrocene conjugates on the levels of ROS and viability of CHO (Chinese Hamster Ovary) - K1 cells. The ROS detection kit and MTT assay were used for this purpose. Treatment of CHO - K1 cells with ferrocene-resveratrol conjugate (RF) resulted in the lowest ROS levels compared to the other compounds tested. Therefore, RF was selected to determine its potential protective effect when CHO - K1 cells were treated with oxidative stress inducers such as tert-butyl hydroperoxide (TBHP) and polychlorinated biphenyl congener - PCB 77. RF significantly reduced 50 μM TBHP-induced ROS levels at all concentrations tested (1; 2,5 and 20 μM). In addition, lower ROS levels were determined in cells pre-incubated with RF and then exposed to 75 μM PCB 77 compared to cells exposed to PCB 77 alone, but the effect was not statistically significant.
