Abstract
Escherichia coli jedan je od najpoznatijih i najvažnijih modelnih mikroorganizama. Zbog dobrog poznavanja genetike i biologije ove bakterije, njezinog kratkog generacijskog vremena te jednostavnih uvjeta uzgoja, koristi se kao domaćin za biotehnološku proizvodnju raznih kemikalija. Razvoj velikog broj tehnika genetičkog inženjerstva bio je usmjeren upravo na ovu korisnu bakteriju, te se stoga danas njezinim genomom može relativno lako manipulirati. Cilj ovog rada je konstruirati plazmid pBAD-RadH, ekspresijski plazmid za bakteriju E. coli na kojem se pod kontrolom inducibilnog araBAD promotora nalazi gen RadH koji kodira za R-specifičnu alkohol-dehidrogenazu. Uspješnost konstrukcije plazmida pBAD-RadH potvrđena je elektroforezom i restrikcijskom analizom.
Abstract (english)
Escherichia coli is one of the best known and most important model microorganisms. Due to a good knowledge of the genetics and biology of this bacterium, its short generation time and simple growing conditions, it was used as a host for the biotechnological production of various chemicals. The development of a large number of genetic engineering techniques was aimed precisely at this useful bacterium, and therefore today its genome can be relatively easily manipulated. The aim of this work is to construct the plasmid pBAD-RadH,which is an expression plasmid for the bacterium E. coli on which, under the control of the inducible araBAD promoter, there is the RadH gene that encodes Rspecific alcohol dehydrogenase. The success of the construction of the plasmid pBAD-RadH was verified by electrophoresis and restriction analysis
