Abstract
Protein IqgC sličan je proteinima IQGAP, a ima svojstva i RasGAP proteina. Prethodno istraživanje je pokazalo da protein IqgC regulira fagocitozu i makropinocitozu deaktivacijom male GTPaze RasG. Cilj ovog rada bio je otkriti ima li protein IqgC ulogu u staničnoj adheziji i ostvaruje li interakciju s malom GTPazom RapA, koja ima ulogu u regulaciji stanične adhezije. Metodama pull-down i koimunoprecipitacije potvrđena je interakcija proteina IqgC s proteinom RapA divljeg tipa, konstitutivno negativnim mutantom S19N i konstitutivno aktivnom formom Q65E. Testom pojedinačnih stanica, u kojem su korištene stanice deficijentne za protein IqgC (iqgC- ), utvrđeno je da protein IqgC sudjeluje u adheziji osjetljivoj na EDTA.
Abstract (english)
IqgC protein shares properties of both IQGAP and RasGAP proteins. It is already known that IqgC is involved in the regulation of macropinocytosis and phagocytosis by deactivating a small GTPase protein RasG . The goals of this paper were to investigate the potential role of IqgC in the regulation of cell adhesion and interaction with the small GTPase RapA. Interaction of the IqgC with wild type, constitutively inactive mutant (S19N) and constitutively active form (Q65E) of RapA was confirmed by using pull-down assay and coimmunoprecipitation assays. The results of the single-cell adhesion assay also implicate IqgC in the regulation of the EDTA-sensitive cell adhesion.
