Abstract
Pojedini Lactiplantibacillus plantarum sojevi sintetiziraju antimikrobne metabolite bakteriocine, čiji aplikativni aspekt obuhvaća primjenu u biokonzerviranju, ali je i potencijalna strategija za smanjenje antibiotičke rezistencije. Cilj ovog diplomskog rada bio je odrediti povezanost fenotipa sinteze bakteriocina s antimikrobnim učinkom L. plantarum sojeva prema Listeria monocytogenes ATCC®19111™ i Staphylococcus aureus 3048. Supernatant kulture L. plantarum sojeva je inhibirao formiranja biofilmova test-mikroorganizama. PCR metodom potvrđena je prisutnost gena plnA, plnEF i plnJ u genomu L. plantarum sojeva. Tijekom kultivacije Lactiplantibacillus sojeva uz određivanje antimikrobne aktivnosti, praćen je bakterijski rast i acidifikacija. Prema rezultatima, sojevi L. plantarum SF15C i L. plantarum D13 iskazuju značajno inhibicijsko djelovanje tijekom stacionarne faze rasta, koje se pripisuje proizvedenim plantaricinima. Gubitak antimikrobnog djelovanja supernatanata kulture pojedinih Lactiplantibacillus sojeva pripisuje se inaktivaciji bakteriocinske aktivnosti. Ovi rezultati su temelj za daljnju izolaciju i karakterizaciju plantaricina koje proizvode L. plantarum D13, s ciljem primjene u fermentacijskim procesima za smanjenje kontaminacija ovim patogenima.
Abstract (english)
Certain Lactiplantibacillus plantarum strains synthesize antimicrobial metabolites bacteriocins, with applications in biopreservation and potential as an alternative antimicrobial strategy. This thesis aimed to examine the correlation between bacteriocin synthesis phenotype and the antimicrobial effect of L. plantarum strains against Listeria monocytogenes ATCC®19111™ and Staphylococcus aureus 3048. The culture supernatant of L. plantarum strains inhibited biofilm formation by the test-microorganisms. PCR analysis confirmed the presence of plnA, plnEF, and plnJ genes in the L. plantarum genome. During cultivation of Lactiplantibacillus strains, bacterial growth, acidification, and antimicrobial activity were monitored. L.plantarum SF15C and D13 strains exhibited a significant inhibitory effect during the stationary growth phase, attributed to plantaricin production. The reduction in antimicrobial activity in the culture supernatants of certain Lactiplantibacillus strains is attributed to bacteriocin inactivation. These findings lay the groundwork for further isolation and characterization of plantaricins produced by L. plantarum D13, facilitating their application in fermentation processes to mitigate contamination.
