Abstract
Prekomjerna i neprikladna uporaba antibiotika pokrenula je razvoj antibiotičke rezistencije u mikrobnim zajednicama. Antibiotici se pri uzgoju životinja ne koriste samo za liječenje infekcija, već izvan Europske unije i kao promotori njihovog rasta, što može uzrokovati kontaminaciju hrane životinjskog podrijetla ostacima antibiotika. Takva hrana može postati vektorom za prijenos antibiotičke rezistencije na humani mikrobiom. Stoga je cilj ovog rada bio razviti metodu za identifikaciju ostataka cefalosporinskih antibiotika u jajima metodom tekućinske kromatografije – tandemske spektrometrije masa (LC-MS/MS) i usporediti dobivene rezultate s rezultatima mikrobiološkog testa za određivanje ostataka antibiotika. Mikrobiološki test ne može se koristiti kao metoda za određivanje ostataka cefalosporinskih antibiotika u jajima zbog nedovoljne osjetljivosti, za razliku od razvijene LC-MS/MS metode koja se prema dobivenim rezultatima pokazala pogodnom za određivanje ostataka cefalosporinskih antibiotika u jajima, osim za desfuroil-ceftiofur.
Abstract (english)
Excessive and inappropriate use of antibiotics has triggered the evolution of antibiotic resistance in microbial communities. When raising animals, antibiotics are not only used to treat infections, but also outside the European Union as promoters of their growth, which can cause contamination of food of animal origin with antibiotic residues. Such food can become a vector for the transfer of antibiotic resistance to the human microbiome. Therefore, the aim of this work was to develop a method for the identification of cephalosporin antibiotic residues in eggs using the method of liquid chromatography - tandem mass spectrometry (LC-MS/MS), to compare and obtain the results with the results of a microbiological test for the determination of antibiotic residues. The microbiological test cannot be used as a method for determining cephalosporin antibiotic residues in eggs due to insufficient sensitivity. On the contrary, according to the obtained results, the developed LC-MS/MS method proved to be suitable for the determination of residues of cephalosporin antibiotics in eggs, except for desfuroyl-ceftiofur.
