Abstract
Modifikacija strukture nativnih peptida s ferocenskim kalupom rezultira povećanom stabilnošću i aktivnošću rezultirajućih peptidomimetika. U ovom je radu provedena in vitro biološka evaluacija homokiralnih konjugata ferocen-1,1'-diamina s L- i D- Val (1, 4), Leu (2, 5) i Phe (3, 6) pri čemu je ispitan njihov utjecaj na morfologiju staničnih kultura HFF-1 i HepG2. Rezultati MTT testa pokazali su najveće antiproliferativno djelovanje ferocenskih konjugata s Phe (3, 6), koje je dovelo do vidnog narušavanja morfologije tumorske stanične linije HepG2 i njezinog odumiranja. Nadalje, jače inhibicijsko djelovanje konjugata 6 na tumorsku staničnu liniju HepG2 u odnosu na zdravu staničnu liniju HFF-1 indicira njegov terapeutski potencijal.
Abstract (english)
Modification of the structure of native peptides with a ferrocene template leads to increased stability and activity of the resulting peptidomimetics. In this study, an in vitro biological evaluation of the homochiral conjugates of ferrocene-1,1'-diamine with L- i D- Val (1, 4), Leu (2, 5) and Phe (3, 6) was performed to investigate their effect on the morphology of HFF1 and HepG2 cell cultures. The results of MTT assay showed the highest antiproliferative activity of the ferrocene conjugates with Phe (3, 6), which caused visible disruption of the morphology and death of the HepG2 tumor cell line. The stronger inhibitory effect of conjugate 6 on the HepG2 tumor cell line compared to the healthy HFF-1 cell line also indicates its therapeutic potential.
