Abstract
U ovom radu istraživana je flokulacijska sposobnost pivskih kvasaca Saccharomyces uvarum WT i Saccharomyces pastorianus NCYC 975 korištenjem Helmovog i Stratfordovog testa. Tijekom 120 sati fermentacije proučavani su različiti parametri vrenja te morfologija i fiziologija stanica pivskog kvasca. Tijekom početna 24 sata fermentacije došlo je do smanjenja ukupne veličine stanica kvasca S. uvarum WT, dok je zatim tijekom 24-60 sata došlo do povećanja stanica. Zatim je ponovno uslijedilo smanjenje veličine stanica sve do završetka fermentacije uz pretpostavku da je do navedene pojave došlo zbog taloženja flokula. S druge strane volumen stanica kvasca S. pastorianus se do 54. sata povećavao, a potom se smanjio do ustaljene vrijednosti. Kultivacija kvasaca uz dodatak 1% (v/v) etanola uzrokovala je smanjenje pojave flokulacije za 40 %. Dodatak 1% (v/v) CaCl2 nije uzrokovao gubitak pojave flokulacije kod S. pastorianus NCYC 975, dok je kod S. uvarum WT primijećeno smanjenje flokulacije za 30 %. Kultivacija kvasaca uz 15 µg/mL cikloheksimid uzrokuje gubitak flokulacije. Dodatak 10 mM EDTA u potpunosti sprječava gubitak flokulacije u prisutnosti glukoze kod oba kvasca. Rezultati dobiveni provođenjem Helmovog testa ne odstupaju značajno od rezultata dobivenih provođenjem Stratfordovog testa. Također dobiveni rezultati potvrđuju da su obje metode prilično precizne za kvantifikaciju pojave flokulacije kod kvasaca. Nadalje, navedeni rezultati također doprinose razjašnjenju uloge nutrijenata na fiziološku kontrolu flokulacije pivskih kvasaca.
Abstract (english)
In this work, the flocculation ability of two brewing yeast strains, Saccharomyces uvarum WT and Saccharomyces pastorianus NCYC 975 has been investigated by means of a Helm's and Stratford's tests. The effects of various fermentation parameters and yeast cell physiology and morphology were studied during 120 h fermentation. Cell size of yeast S. uvarum WT initially decreased during the first 24 h of fermentation then increased during 24-60 h fermentation period. Cell size subsequently declined until the end of fermentation presumably due to the floc settling. On the other hand, cell size of yeast S. pastorianus NCYC 975 during the first 54 hours increased and then decreased to constant value. The incubation of yeasts in 1% (v/v) ethanol induce 40% flocculation loss, while 1% (v/v) CaCl2 did not induce flocculation loss in S. pastorianus NCYC 975, but 30% loss in S. uvarum WT. Incubation of cells in the presence of 15 µg/mL cycloheximide caused flocculation loss. The presence of 10 mM EDTA prevented completely the loss of the flocculation in the presence of glucose. The results obtained with the Helm’s test do not differ significantly from those obtained with Stratford’s test. The results presented clearly demonstrated that the both methods are accurate for quantification of brewing yeast flocculation. The results contribute to the elucidation of the role of nutrients on the physiological control of brewing yeast flocculation.
