Abstract
During this training, we studied interaction of 5-HT6 receptor with β-arrestins and downstream signalling of 5-HT6R, on ERK and mTOR signalling pathway. We provided first evidence of interaction of 5-HT6R with β-arrestins 2 and demonstrated that WAY-208 466 and 5-HT act as agonists on this recruitment, SB-271 act as antagonist and SGS-518 act as inverse agonist. AlphaScreen results from NG 108-15 lysates silenced for β-arrestin 2 revealed decrease in quantity of phosphorylated ERK, leading to conclusion that 5-HT6R could trigger ERK signalling pathway directly through its interactions with β-arrestin 2. We couldn’t obtain any results demonstrating involment of Nf1 on ERK signalling pathway. However we demonstrated involvement of Nf1 in mTOR signalling pathway by westren blot analysis on brain lysates and immunohistochemistry studies on brain sections.
Abstract (french)
Durant ce stage, nous avons étudié l'interaction de R5-HT6 avec les ß-arrestins et leur rôle sur la signalisation de R5-HT6, sur les voies ERK et mTOR. Nous avons montré pour la première fois une intéraction spécifique entre le R5-HT6R et les arrestin 2 et démontré que le WAY 208466 et la 5-HT en favorisant le recrutement de la arrestin 2 agissent comme des agonistes, alors que le SB271 agit comme un antagoniste et le SGS-518 comme un agoniste inverse. La sous-expression des ß-arrestin 2 induit une diminution de la phosphorylation de ERK induite par l'activation du récepteur et mesurée par dosage AlphaScreen, ce qui suggère que la stimulation de ERK implique l'intéraction des ß-arrestin 2. Nous n'avons pas réussi à mettre en évidence un rôle de Nf1 sur l'activation de la voie ERK induite par les agonistes. Par contre, nous montrons qu'une sous expression de Nf1 chez des souris module la voie mTOR aussi bien sur des lysats de tissus cérébraux que sur coupes de cerveau.
