Abstract
Antibiotici se koriste za liječenje bakterijskih infekcija kod ljudi i životinja. Njihovo učestalo i nepotrebno korištenje prouzročilo je pojavu rezistentnih patogenih bakterija. Poznavanje mehanizama djelovanja antibiotika te mehanizama rezistencije na antibiotike postaje neophodna za suzbijanje i kontrolu rezistentnih bakterija. S ciljem razvitka brze metodologije za određivanje mehanizama rezistencije, u ovom radu testirana je nova metoda bazirana na proteomskoj analizi patogene bakterije. Uropatogena bakterija Escherichia coli MFBF 10519 pokazala je rezistentnost na šest različitih β-laktamskih antibiotika te je korištena kao test mikroorganizam za ispitivanje mehanizama rezistencije metodom CAF-/CAF+. Rezultati razlikovnog proteomskog ispitivanja mehanizama rezistencije bakterije E. coli MFBF 10519 na penicilin G ukazuju na povećanu sintezu osam proteina. Četiri od njih, β-laktamaza LEN-26, porin OmpX, transposaza IS66 i protein OmpA, direktno utječu na rezistenciju E. coli MFBF 10519 na penicilin G.
Abstract (english)
Antibiotics are used for the treatment of bacterial infectious diseases of people and animals. High prevalence of antibiotic use and abuse has led to pathogenic bacteria resistance development. Understanding the antibiotic mode of action and bacterial resistance mechanism becomes essential to infection moderation and regulation. In order to develop a fast method for resistance mechanism identification, in this work we tested a new method based on proteomic analysis of the whole proteoma. Uropathogenic bacterium Escherichia coli MFBF 10519 which showed resistance to 6 different β-lactamic antibiotics was used as a test microorganism for resistance mechanism identification and CAF-/CAF+ method was used. Results a distinctive proteomics testing for resistance mechanism in bacterium E. coli MFBF 10519 to Penicillin G have shown 8 different protein expressions. Four of these proteins β-lactamase, porin OmpA, transposase IS66 and protein OmpA have a role in the resistance to Penicillin G.
