Abstract
Zbog sve veće pojave rezistencije bakterija na postojeće lijekove, do koje je došlo zbog pretjerane i neutemeljene upotrebe trenutno dostupnih antibiotika, u zadnjih se desetak godina pojavila potreba za pronalaskom novih antibiotika. Antibiotici spadaju u skupinu spojeva koji se nazivaju sekundarni metaboliti, a čiju veliku većinu čine poliketidi i neribosomalno sintetizirani polipeptidi. Takve sekundarne metabolite uglavnom proizvode bakterije roda Streptomyces, a služe im kao kompetitivna prednost u odnosu na druge bakterije. Međutim, problem u pronalaženju novih antibiotika je što ih većina nije eksprimirana u mjeri da bi mogla biti detektirana i okarakterizirana standardnim znanstvenim metodama. Cilj ovog diplomskog rada je bilo anotiranje genoma slabo poznate bakterijske vrste Streptomyces sp. C bioinformatičkim programskim paketom ClustScan kako bi se pronašli geni koji kodiraju za enzime za sintezu potencijalno novih antibiotika nepoznatih struktura.
Abstract (english)
Due to increased bacterial resistence to existing drugs, which was caused by an excessive and unjustified use of currently available antibiotics, in the last several years a need for new novel antibiotics arose. Antibiotics are a group of compounds which, among others, are called secondary metabolites and mostly consist of polyketides and nonribosomally synthesized peptides. These secondary metabolites are mostly produced by bacteria of the Streptomyces genus and serve to their producers as a competitive advantage relative to other bacteria. The problem in finding new antibiotics is that most of them are not expressed to the extent of being detected and characterized by standard scientific methods. The goal of this thesis is to annotate the genome of a not well known species of bacteria, Streptomyces sp. C using ClustScan, the bioinformatics computer program, in order to find genes which encode for enzymes which produce potentially new antibiotics of unknown structures.
