Abstract
Cilj ovog rada je determinirati faze mitoze u meristemu korijena klijanaca pšenice. U prvoj fazi istraživanja je bilo potrebno naklijati zrna pšenice. U drugoj fazi su metodom gnječenja napravljeni preparati stanica korijena pšenice. U trećoj fazi su pod mikroskopom određene faze mitoze i računao se mitotski indeks. Za mitotski indeks bilo je potrebno izbrojati 500 stanica te razlučiti stanice u interfazi od onih u nekoj od faza diobe. Detalji morfologije kromosoma nisu vidljivi zbog preskočenog tretmana fiksacije te razlika između interfaze i profaze nije bila dovoljno jasna. Samu vrijednost mitotskog indeksa dobivenu ovim istraživanjem zato ne treba uzeti u obzir.
Abstract (english)
Purpose of this work is to determinate stages of mitosis in apical meristem of wheat. In first stage of research wheat seeds are placed to germinate. In second stage squash method is used to make cell section of wheat root. Third stage considered microscopic observing, assessment of stages in which cell is found and calculation of mitotic index. Morphologic details of chromosomes were not clearly visible and it was hard to distinguish difference between interphase and prophase. Value of mitotic index should not be considered as right.
