Abstract
U ovom radu provedena je izolacija genomske DNA iz listova soje i suncokreta, a u istraživanju korištene su CTAB i SDS metode izolacije. Cilj rada bio je izolirati DNA te utvrditi čistoću i količinu izolirane DNA. Iz obje ispitivane biljne vrste uspješno je izolirana DNA optimalnih vrijednosti omjera absorbanci (A260/280 1,7 – 2,00; A260/230 >2,0). Najveća količina DNA izolirana je CTAB (1225,6 ng/μl), dok je najmanja količina izolirana SDS metodom (258,51 ng/μl).
Abstract (english)
In this work, genomic DNA isolation from soybean and sunflower leaves was performed, and using CTAB and SDS isolation methods. The aim of the study was to isolate DNA and determine its purity quantity.DNA was successfully isolated from both plant species with optimal absorbance ratio (A260/280 1,7 – 2,00; A260/230 >2,0). The highest DNA yield was recorded with CTAB (1225.6 ng/μl) , while the lowest yield was recorded with SDS method (258.51 ng/μl).
