Abstract
Neuropeptid Y (NPY) je peptid koji se sastoji od 36 aminokiselina. U posljednjih nekoliko godina ustanovljeno je da je NPY važan ko-transmiter u perifernom živčanom sustavu. U ovom radu dan je pregled literaturnih podataka o kardiovaskularnim bolestima, lijekovima koji se koriste u njihovom liječenju i ulozi estrogena. Detaljnije su opisane ishemijska bolest srca i hipertenzija jer se smatra da će NPY antagonisti biti korisni u njihovom liječenju, budući da otpušteni NPY uzrokuje konstrikciju krvnih žila i smanjenje srčanog tonusa. U svrhu daljnjeg istraživanja potencijalnog učinka na kardiovaskularne bolesti sintetiziran je hidroklorid amida (R)-N 2 -(difenilacetil)-(R)-N-[1-(4- metoksifenil)etil]arginin (7), a njegova struktura je dokazana spektroskopskim metodama (1H i 13C NMR). U daljnjim istraživanjima, koja prelaze okvire ovoga diplomskog rada, bit će ispitano njegovo djelovanje na kardiovaskularni sustav
Abstract (english)
Neuropeptide Y (NPY) is a peptide consisting of 36 aminoacids. Recently, it has been established that NPY is an important co-transmitter in peripheral nervous system. This thesis gives an overview of the current literature regarding cardiovascular diseases, drugs used in cardiovascular medicine and the role of estrogen. Myocardial ishemic heart disease and hypertension are described in more detail since NPY antagonists might be useful in the treatment of these cardiovascular diseases because NPY causes constriction of blood vessels in both the heart and it reduces the cardiac vagal tone. In this work we have prepared (R)-N 2 -(diphenylacetyl)-(R)-N-[1-(4- hydroxyphenyl)ethyl]arginine amide hydrochloride (7), NPY antagonist, which was characterized by standard spectroscopic methods (1H and 13C NMR). Future investigations will focus on the evaluation of its role in cardiovascular diseases.
