Abstract
Egzosomi su izvanstanične mikrovezikule koje izlučuju gotovo sve stanice u organizmu. Mogu se pronaći u većini tjelesnih tekućina kao što su urin, krv, slina, cerebralna tekućina i majčino mlijeko. Njihov sadržaj usko korelira sa sadržajem stanice iz koje potječu, a s obzirom da ih između ostalog izlučuju i stanice nastale patološkim procesima, odnosno tumorske stanice, njihova izolacija i daljnje promatranje igra veliku ulogu u ranoj dijagnostici tumorskih bolesti. Cilj ovog diplomskog rada bio je usporediti dvije metode izolacije i doći do zaključka koja je od njih prikladnija za izolaciju egzosoma. Prva metoda je bila metoda centrifugiranja u kojoj se izolacija egzosoma temelji na njihovoj precipitaciji pri jačim snagama centrifugalnog polja. Nadalje, korištena je metoda koja za poboljšanju precipitaciju egzosoma koristi komercijalno dostupan precipitacijski polimer. Nakon analize uzoraka na Zeta-sizeru može se zaključit da je metoda s precipitacijskim polimerom uspješnija u izolaciji egzosoma. Također, analizirani su i uvjeti skladištenja. Iz rezultata analize zaključeno je da uzorak izoliranih egzosoma treba biti svježe pripravljen prije daljnje analize veličine čestica.
Abstract (english)
Exosomes are extracellular microvesicules which are produced by almost every cell in human organism. They can be found in most body fluids such as urine, blood, saliva, celebral fluid and breast milk. Exosome content closely correlates with the content of the cell they originate from, and as they are also produced by tumour cells, their isolation and further examination is greatly important in early diagnosis of tumour illnesses. The goal of this research was to compare two methods of exosome isolation and conclude which one of them is more suitable to be used for isolation of exosomes. First method used was the method of centrifugation in which the isolation of exosomes is based on their precipitation by centrifuge forces. Furthermore, other method used commercially available kit, which contains special polimer for exosome precipitation. After sample analysis on Zeta-sizer it could be concluded that the method using TEI reagent is much more successful in exosome isolation. The storage conditions were also analysed. From the results oft he analyses it is concluded that the exosomes should be analysed freshly after isolation in order to get optimal results.
