Abstract
Sve veći broj istraživanja potvrđuje neuroprotektivno djelovanje GLP-1 i njegovih analoga aktivacijom GLP-1 receptora (GLP-1R) u središnjem živčanom sustavu. Iako je takvo djelovanje potencijalno značajno za terapiju neurodegenerativnih bolesti, točan mehanizam i signalizacija GLP-1R u mozgu još uvijek nisu u potpunosti istraženi. U ovom istraživanju, ispitane su akutne promjene u ekspresiji markera neuronske aktivnosti (c-fosa), metaboličke aktivnosti (inzulinskog receptora, IR) i apoptoze (pERK, p70S6k, PDK1) u hipokampusu (HPC) i hipotalamusu (HPT) mozga zdravih štakora, nakon intracerebroventrikularne (icv) primjene različitih doza antagonista GLP-1 receptora, eksendina 9 (Ex-9). Rezultati su analizirani faktorskom analizom mješovitih podataka i prikazani kao apsolutne promjene u ekspresiji pojedinog proteina te su izrađene matrice korelacije promjena u ekspresiji proteina svake skupine u odnosu na kontrolu. Napravljena je 3D vizualizacija kao dinamički prikaz promjena u ekspresiji proteina kod različitih doza u odnosu na kontrolu. Naši rezultati pokazuju pad neuronske i metaboličke aktivnosti, a porast apoptoze u HPC i HPT, povećanjem doze Ex-9. Primijećena je konzistencija u biokemijskim promjenama unutar svake regije zasebno, ali i razlike u promjenama koje se događaju u jednoj regiji u odnosu na drugu. Potrebna su dodatna istraživanja kako bi se bolje objasnile razlike u biokemijskim promjena koje se događaju u HPC u odnosu na HPT.
Abstract (english)
Increasing numbers of research confirm the neuroprotective effects of GLP-1 through the activation of GLP-1 receptors (GLP-1R) in the central nervous system. Although the effects of GLP-1 could prove significant in treatment of neuro-degenerative diseases, the exact mechanism and signalization of GLP-1R in the brain have not been completely uncovered. This research tests the acute changes in marker expressions of neural activity (c-fos) along with metabolic activities (insulin receptor, IR) and apoptosis (pERK, p70S6k, PDK1) inside the hippocampus (HPC) and hypothalamus (HPT) regions of healthy rat brains after the intracerebroventricular (icv) administration of various doses containing antagonists of the GLP-1 receptor, exendin 9 (Ex-9). Factor analysis of mixed data and correlation matrix were made and the results are shown as absolute changes in protein expression. The 3D visualization has been constructed as a dynamic display of changes in protein expression after the administration of various doses of Ex-9. Our results show a decrease of neural and metabolic activity along with an increase of apoptosis inside the HPC and HPT with the increase of Ex-9. A consistency of biochemical changes through every individual region has been noted along with the difference of changes present in one region in relation to the other region. Further research would be necessary to better explain the differences in biochemical changes present in the HPC compared to the HPT.
