Abstract | Vrste koje pripadaju rodu Petasites su višegodišnje biljke s velikim listovima koji se razvijaju nakon cvatnje i debelim korijenom. U ovo istraživanje bile su uključene sljedeće vrste: Petasites albus (L.) Gaertner, P hybridus (L.) P. Gaertner, B. Meyer & Scherb., P. paradoxus (Retz.) Baumg., P. kablikianus Tausch ex Berchtold. Budući da je danas poznato da oksidacijski stres pridonosi razvoju kroničnih degenerativnih bolesti, razvoju raka i starenju, a fenoli i flavonoidi su kemijski spojevi kojima se pridaje sve više pažnje kao potencijalnim antioksidansima, cilj ovog istraživanja bio je spektrofotometrijski odrediti i usporediti sadržaj flavonoida i ukupnih fenola te antioksidativni učinak između različitih vrsta roda Petasites, kao i između istih vrsta (P. albus i P. hybridus) sakupljenih na različitim lokacijama. Sadržaj flavonoida se odredio metodom koja se temelji na njihovom keliranju s AlCl3 pri čemu nastaju žuto obojeni kompleksi, a kretao se između 2,21 ± 0,09 i 5,59 ± 0,05 mg ekvivalenta kvercetina/g uzorka. Sadržaj ukupnih fenola odredio se pomoću Folin-Ciocalteuovog reagensa te se kretao od 4,43 ± 0,09 do 19,92 ± 2,90 mg ekvivalenta galne kiseline/g uzorka. Antioksidativni učinak odredio se pomoću slobodnog radikala 2,2-difenil-1-pikrilhidrazila (DPPH) te se kretao od 11,78 ± 1,34 mg do 41,98 ± 0,12 mg ekvivalenta Troloxa/g suhog biljnog materijala. Statistička obrada pokazala je vrlo dobru pozitivnu povezanost između sadržaja ukupnih fenola i rezultata mjerenja antioksidativnog učinka (r = 0,93, p < 0,001). S obzirom na dobivene rezultate ove vrste mogle bi imati potencijal kao antioksidansi te sudjelovati u prevenciji kroničnih degenerativnih bolesti i raka. |
Abstract (english) | Species belonging to the genus Petasites are perennial herbs with large leaves that develop after flowering and thick rhizomes. Species included in this study were: Petasites albus (L.) Gaertner, P. hybridus (L.) P. Gaertner, B. Meyer & Scherb., P. paradoxus (Retz.) Baumg., P. kablikianus Tausch ex Berchtold. Since it is known today that oxidative stress contributes to the development of chronic degenerative diseases, cancer and aging and phenols and flavonoids are chemical compounds which are given increased attention as potential antioxidants, the aim of this study was to determine and compare total phenolic and flavonoid content as well as antioxidant activity between different species of the genus Petasites and also between the same species (P.albus and P. hybridus) collected on different locations. Flavonoid content was determined by a method based on their chelating ability with AlCl3 where they form a yellow coloured complex and it ranged from 2.21 ± 0.09 to 5.59 ± 0.05 mg quercetin equivalent/g sample. Total phenolic content was determined by using the Folin-Ciocalteu's reagent and ranged from 4.43 ± 0.09 to 19.92 ± 2.90 mg gallic acid equivalent/g sample. Antioxidant activity was determined by using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) reagent and ranged from 11.78 ± 1.34 to 41.98 ± 0.12 mg Trolox equivalent/g dry plant material. .Statistical analysis showed very good positive correlation between total phenolic content and results of antioxidant activity measurements (r = 0.93, p < 0.001). Considering the obtained results these species might have potential as antioxidants and could contribute to prevention of chronic degenerative diseases and cancer. |
