Urine is waste product of the body but it is very important for the diagnosis of many diseases. Qualitative analysis of urine includes the organoleptic and chemical analysis and examination of the urinary sediment. Vacuum tubes have been recently implemented into the routine practice. The aim of this study was to investigate whether the composition of urine changes if instead of pouring out of the cup in a test tube we use vacuum tubes. This study was done in Clinical Institute of Chemistry at the University Hospital Center "SESTRE MILOSRDNICE". We have analyzed a series of first morning urine samples (N=163) received from Daily hospital of the Clinical Institute of Nephrology and Dialysis. For sampling were used urine collection cups and vacuum tube VACUETTE® of Greiner Bio-One International (Kremsmünster, Austria). Each sample was mixed and poured out of the cup in a test tube and drawn by vacuum tube. Chemical analysis was performed using the test strip Combur10 TEST M and for the sediment samples were centrifuged 10 min at 1500 rpm. Sediment was observed by light microscopy. Results of routine urinalysis were statistically analyzed, calculating the Kappa coefficient and Chi-square test. There was no statistically significant difference between samples. Based on our results, we conclude that the vacuum does not impair the elements of sediment or change the composition of the sample and can be used in routine. The use of vacuum tubes standardizes volume (10,5 ml) that enters the analysis of the sediment. Sampling is simplified and medical staff is protected from potentially infectious samples.
