Abstract | Gemcitabin (2', 2'-difluorodeoksicitidin, dFdC) je antitumorski lijek koji je analog pirimidina. Indiciran je kod karcinoma gušterače, karcinoma pluća nemalih stanica, karcinoma mokraćnog mjehura, jajnika, sarkoma mekih tkiva i non-Hodgkinovih limfoma.
Za analizu gemcitabina i njegovog onečišćenja citozina odabrana je micelarna elektrokinetička kromatografija. U ovom radu optimizirane su vrste i koncentracije organskog otapala (acetonitril, etanol i metanol) koji se dodavani u otopinu radnog pufera. Ispitan je dodatak neinskog surfaktanta Tritona s ciljem postizanja simetričnih pikova gemcitabina i citozina.
Optimalni uvjeti analize su postignuti pri 20 mM fosfatnog pufera pH 7,0, 70 mM SDS-a, 5 % (v/v) metanola, primijenjenom naponu od 25 kV, 25 ºC, detekciji pri 275 nm, hidrodinamičkom injektiranju 50 mbar, 6 s. Pri navedenim uvjetima postignuto je razlučivanje između pikova gemcitabina i citozina od 0,71.
Predmet daljnjeg istraživanja bit će modifikacija uvjeta analize dodatkom aditiva kojima bi postiglo bolje razlučivanje, bolji oblik pikova i veća simetričnost pikova. |
Abstract (english) | Gemcitabine (2', 2'-difluorodeoxycytidine, dFdC) is an anticancer drug which is a pyrimidine analog. It is used in treating pancreatic cancer, non-small cell lung cancer, bladder cancer, ovarian cancer, soft tissue sarcomas and non-Hodgkin limfoma.
Micellar electrokinetic chromatographic approach was chosen to develop a method which would be able to separate gemcitabine and its impurity cytosine. In this research, different types and concentrations of organic solvents (acetonitrile, ethanol and methanol), which were added to the working buffer solution, were optimized. Triton, a neutral surfactant, was tested with the aim of achieving symmetric peaks of gemcitabine and cytosine.
The optimum analysis conditions were achieved with 20 mM phosphate buffer pH 7,0, 70 mM SDS, 5 % (w/w) methanol, applied voltage of 25 kV, at 25 ºC, detection at 275 nm and hydrodynamical injection of 50 mbar, 6s. The resolution of gemcitabine and cytosine peaks achieved under these conditions was 0,71.
The subject of further research will be to modify the conditions of analysis by adding additives to achieve better resolution, better shape of the peaks and greater symmetry. |
